ObjectiveTo investigate the relationship between the expression of survivin and the methylation of CpG island in survivin gene exon 1 region,and to explore the regulatory mechanism of survivin gene in CA tissues.Methods(1)The expression of survivin were examined by immunohistochemistry in paraffin-embedded tissues from 30 cases of CA lesion,10 cases of normal cervical and 10 cases of normal foreskin.(2)Genomic DNA and total RNA were isolated;(3)RT-PCR approach was used to detect the mRNA level of survivin;(4)Methylation specific PCR(MSP) technique were used to analyze the methylation status of the CpG island in the survivin gene exon 1 region.Results(1) 27 cases of the 30 condyloma acuminatum(90%) expressed survivin protein.In contrast,in 20 normal tissues,only 1 cases of normal cervical tissues(5%)expressed survivin.The survivin expression was signficantly greater in the CA than in normal tissues(P=0.000).(2)Survivin mRNA was signficantly higher in lesions of CA(28/30, 93.3%) than in normal tissues(1/20,5%)(P=0.000).(3)The CpG sites in the survivin gene exon 1 region were mostly unmethylated in CA tissues(26/30,86.7%) compared with normal tissues(3/20,15%),there was a significantly statistical difference between the two groups(P=0.001).(4) There is a correlation between the status of unmethylated CpG island in survivin exon 1 region and survivin mRNA expression(P =0.014).ConclusionThe expression of survivin in CA tissues might be regulated by the unmethylation of CpG island in surviving exon 1 region,and it maybe play a role in CA pathogenesis.
