Objective:1.To investigate the expression of HtrA2/Omi and XIAP protein in colorectal cancer tissues.2.To investigate the correlation of HtrA2/Omi and XIAP protein and with clinicopathologic factors.3.To investigate the role of HtrA2/Omi and XIAP protein in the occurrence and prognosis of colorectal cancer,and to evaluate their value for prognosis of the patients with colorectal cancer.Methods:1.To collect 70 primary colorectal cancer cases and 20 colorectal adenoma cases during July 1,2001 and July 1,2003 in cancer hospital of Hunan Province,which were all treatment by operation and confirmed by pathology results and their paraffin-embedded specimens were saved very well.2.Immunohistochemical technique was used to detected the expression of HtrA2/Omi and XIAP protein in the paraffin-embedded specimens of 70 colorectal cancer cases and mucosa adjacent to colorectal cancer and 20 colorectal adenoma cases.3.The software package spss 13.0 was used for statistical analysis.Result:1.The high expression rate of HtrA2/Omi protein was 57.14%in 70 colorectal cancer samples and 78.57%in normal controls of colorectal mucosal tissues,while 60.00%was in 20 colorectal adenoma tissues. There was a significant difference between three groups(p<0.05).2.The positive expression rate of XIAP was 50.00%in 70 colorectal cancer samples and 14.29%in normal controls of colorectal mucosal tissues,while 35.00%was in 20 colorectal adenoma tissues.There was a significant difference between three groups(p<0.05).3.By statistical analysis,the expression of HtrA2/Omi was found to be associated with different types of histology of colorectal cancer:The high expression rate of poorly differentiated adenocarcinoma (25.00%) was significantly lower than moderate adenocarcinoma (68.80%)(p<0.05).The well-differentiated adenocarcinoma was 50.00%,and no different with other groups(p>0.05)4.By statistical analysis,there was no correlation of XIAP protein with the clinicopathologic factors such as age,sex,location,infiltration,lymph node metastasis and Dukes stage(p>0.05).5.By Spearman analysis,HtrA2/Omi expression was not associated with XIAP protein expression in colorectal cancer.The correlation coefficient was-0.231(p=0.054).6.The total five-year survival rate was 76.00%in 70 cases of patients. The five-year survival rate after surgery in the patients with high expression rate of HtrA2/Omi(82.50%)was significantly higher than the low one(70.00%).The five-year survival rate after surgery in XIAP expression positive patients was 65.70%,compared with 88.60%survival rate in XIAP expression negative patients,which had a significant difference(p<0.05).Multivariate analysis showed that it was lymph node metastasis but not HtrA2/Omi and XIAP was an independent prognostic factor for patients with colorectal cancer.Conclusion:1.The abnormal expression of HtrA2/Omi protein and XIAP protein in colorectal cancer tissues may play a role in the occurrence and development of colorectal cancer.2.Lymph node metastasis was an independent prognostic factor for patients with colorectal cancer,but HtrA2/Omi and XIAP were not.
