| [Background and objective]At present, the bladder carcinoma attack rate is the 8th place in the carcinoma of all over the body in male, and the 12th place in female in china. But the attack rate in our country is less than in western countries. The treatment of bladder carcinoma include operation and irrigation of bladder chemotherapy , but the relapse of bladder carcinoma usually happen after all of the treatments we did . So early diagnosis, relapse and how to treat are the tough problems of bladder carcinoma. Therefore, it is important to investigate some mechanisms of bladder carcinoma. Recently, the research of centrosome is a hot spot after cell cycle, signal conduction and apoptosis about mechanisms of bladder carcinoma.Aurora protein kinases educe a important effect in caryocinesis. Aurora protein kinases include Aurora-A,Aurora-B,Aurora-C. Aurora-A plays important roles in the completion of essential mitotic events such as centrosome separation, spindle assembly and spindle maintenance. Now, Aurora-A is confirmed as an oncogen. it can cause centrosome amplification, chromosome instability and anorthoploid formation to result in tumor happened.To explore mechanism of bladder carcinoma and the effect that Aurora-A educe in bladder carcinoma ,we focused on the expression ofAurora-A protein in bladder carcinoma samples and the normal bladdertissue.[Materials and Methods]40 bladder carcinoma samples that diagnosis by xiangya hospital and 20 normal bladder tissues were collected. , Aurora-A protein expression and location was analyzed with immunohistochemical approach. In forty bladder carcinoma samples there is 35 males and 5 females, age from 19 to 85 years old, average 59 years old. I :12 instances, II :25 instancesIII:3 instances in WHO/ISUP(1998).[Result]1 .Protein of 40 bladder carcinoma samples and 20 normal bladder tissues were collected and assayed for Aurora-A expression by immunohistochemical approach. Among 40 human bladder carcinoma samples, strong cytoplasmic staining (+or++) of Aurora-A protein was detected in 29 tumors, to seize 72.5% .In contrast, only one sample exhibited positive staining(+) for Aurora-A protein among 20 normal adjacent tissues, P<0.01.2.Among 20 non-muscular-invasion bladder carcinoma samples, negative staining (-or±) of Aurora-A protein was detected in 9 samples, positive staining (+or++) have 11 samples; In contrast, among 20 muscular-invasion bladder carcinoma samples, there is 2 and 18 in negative and positive staining, P<0.05 . there is 9 and 3 in negative and positive in Grade I; 5 and 18 in Grade II; 0 and 5 in Grade III. P<0.05[Conclusion]1.Because the expression of Aurora-A in bladder carcinoma samples is different from that in normal bladder tissue, maybe the amplification of Aurora-A is related to the bladder carcinogenesis.2.Over-expression of Aurora-A is detected in bladder carcinoma tissues and correlates with the diferentiation and invasion of bladder carcinoma. It may be considered as a candidate molecular parameter indicating the tumor invasion and prognosis of bladder carcinoma3. Over-expression of Aurora-A may be the mechanisms of bladder carcinoma, so we can treat the bladder carcinoma by depressing the expression of Aurora-A in the future. |
