BACKGROUND:In China and even in the world,bladder cancer is one of the most common malignant tumors in urinary system tumors.The occurrence of bladd er cancer is a complex,multi-factor,multi-step process.We all don't clear the reason.In our working,we use TNM stage to choose its treatment and prognosis.Accroding to the tumor's depth of invasion,we classify it into non-muscle invasive bladder c ancer(NMIBC)and muscular invasive bladder cancer(MIBC).We also refer to the former as superficial bladder cancer.It mainly refers to non-muscle invasive papilla ry carcinoma and tumors that infiltrate into the subcutaneous connective tissue but n ot the muscular layer,ie,Ta and Tl;the latter refers to Bladder neoplasms that infi ltrate into the muscle and underlying tissues,ie T2,T3,and T4.At present,its also plays an extremely important role in the clinical treatment.It determines the devel opment of the treatment,surgical and prognosis for bladder cancer patients by urolo gists.The reason why this scheme was established is mainly due to the difference i n the recurrence rate and survival rate between non-muscle-invasive bladder cancer and muscular invasive bladder cancer.The prognosis of the latter is worse than the former,which seriously reduces the quality and survival time for the patient's life.It is well-known that oncogenes are the most important factor leading to the develo pment of tumors,so reducing the activity of oncogenes and blocking their associat ed signaling pathways will be an important step for our fight against cancer.Object ive:This time we will use R language and bio informatics knowledge to analyze an d excavate differential genes and signaling pathways of muscular invasive and non-muscle-invasive bladder cancer.It will find the prognosis renson of muscular invasiv e bladder cancer.Poor target genes,which will further complement the signal pathw ays in the development of bladder cancer,and provide new ideas for many research ers.At the same time lay the foundation for further research on the muscle-invasive bladder cancer.Methods:First,we screen and download the GeneChip GSE37317 dataset from the PubMed GEO database(https://www.ncbi.nlm.nih.gov/pubmed/),and completed pre-process data by the R language platform which including data entry,quality control,background correction,standardization,and summaries.Then screening the significant differentially expressed genes between muscle invasive bladder canc er and non-muscle invasive bladder cancer,and Hierarchical clustering analysis for t he results.Then use the DAVID website(https://david.ncifcrf.gov/)to annotation gen e function.On the other hand,use the gene ontology KEGG pathway enrichment to analysis of the differential genes.At last,constructed a protein-protein interaction(PPI)network by the STRING website(https://string-db.org/)and Cytoscape software and screened the representative gene modules as the target genes in this study.Re sults:This study showed that there were 145 differentially expressed genes in musc ul invasive bladder cancer and non-muscle invasive bladder cancer,of which 57 gen es were highly expressed and 88 genes were lowly expressed.GO analysis of these differentially expressed genes is mainly involved in extracellular matrix organization,response to mechanical stimuli,collagen catabolism,prostate gland epithelial morph ogenesis,negative regulation of angiogenesis,extracellular matrix decomposition in t he top 10 biological processes and the negative regulation of endothelial cell migrat ion and other processes;KEGG enrichment analysis results show that the relevant signaling pathways include ECM-receptor interaction pathway,amoeba pathway,rheum atoid arthritis,lysosomal pathway,local adhesion pathway.Twenty-six genes with sig nificant differences were selected from the PPI network,17 genes were up-regulated genes and 9 were down-regulated genes.And have six genes participated nine biol ogical processes in the top 10 go term.Conclusion:We have found six genes with significant differential expression may play an important role in the development of muscular bladder cancer.They may be used for targeted therapy of muscular invas ive bladder cancer or as the tumor marker to screening and diagnosis of bladder cancer.
