Objective:1 Effect of glutamine on PPAR-γexpression in lung tissiue of sepsis induced acute lung injury;2:To investigate the anti-inflammatory menchanism of glutamine in sepsis-induced acute lung injury with glutamine,GW9662(an antagonist of PPAR-γ) and combination of these two agents.Methods:Rats were randomly divided into five groups as follow: sham-operated group received 10%dimethylsulfoxide(DMSO) 30min Prior sham-operation,CLP group and C LP+GLN group were received 10%dimethylsulfoxide(DMSO) 30min Prior cecal ligation and puncture operation,GW +CLP+GLN group,GW+CLP gr oup were received the selective PPAR-γantagonist GW9662(1mg/kg intravenously) 30min Prior cecal ligation and puncture operation,glutamine was administered to CLP+GLN group,GW+CLP+GLN group 1 h after cecal ligation and puncture(CLP),saline was administered SHAM group and CLP group 1 h after operation.PPAR-γexpression were evaluated in lung tissue 24 h post-CLP,plasma and lung tissue TNF-αand IL-6 cytokines were assayed by ELISA,last,lung histopathology for occurrence of ALI and lung wet/dry weight ratio were examined.Results:1 The expression of PPAR-γwas significantly lower in CLP group than sham group(P<0.05),and was higher in CLP+GLN group than CLP group(P<0.05);2 PaO2 was significantly higher in sham group than in CLP group,CLP+GLN group,GW+CLP+GLN group and GW+CLP group(P<0.05),PaO2 was significantly higher in CLP+GLN group than CLP group(P<0.05),and was significantly lower in GW+CLP+GLN group than CLP+GLN group(P<0.05),lung wet/dry weight ratio,lung injury score,the concentrations of TNF-α, IL-6,were significantly lower in CLP+GLN group than CLP group (P<0.05),and were significantly higher in GW+CLP+GLN group than CLP+GLN group(P<0.05)Conclusions:Glutamine attenuates inflammatory cytokine release,and prevents acute lung injury following sepsis through PPAR-γup-regulation.
