Objective:To study the relationship of serum concentration of UN,Cr and UA and those in saliva.To investigate the factors influencing the measurement of UN,Cr and UA in saliva,and evaluate the feasibility of the methods for quality control of the measurement of UN,Cr and UA in saliva.Meanwhile,to install the normal reference value of salivary concentration of UN,Cr and UA in our laboratory in order to serve for clinic and identify the value of salivary UN,Cr and UA in diagnosis of certain diseases,Methods:1.Saliva was Collected in 20 healthy individuals by means of citric acid swab stimulus method and citric acid droping in stimulus method.The concentration of UN,Cr and UA in saliva were measured,and the results obtained from the two different collecting methods were compared.2.Saliva was collected before and after gargling in 20 healthy individuals by means of citric acid swab stimulus method and citric acid droping in stimulus method.The concentration of UN,Cr and UA in saliva were measured,The influence of gargling on the salivary levels of UN,Cr and UA were evaluated.3.Saliva was collected before and after lunch in 20 healthy individuals by means of citric acid swab stimulus method.The concentration of UN,Cr and UA in saliva were measured,The influence of dining on the salivary levels of UN,Cr and UA were evaluated.4.Saliva was collected in 20 healthy individuals by means of citric acid swab stimulus method at 6 Am,7 Am,8 Am and 12 Am,as well as 1Pm and 4Pm,The concentration of UN,Cr and UA in saliva were measured,and find out the excreting axiom of salivary UN,Cr and UA during the day and the optimal time for collection of saliva.5.Repeatability determination:Saliva was collected after gargling in 20 healthy individuals by means of citric acid swab stimulus method,The specimen were subpackaged into 6 tubes respectively and stored the samples in -20℃,The concentration of UN,Cr and UA of one sample was measured in 6 continuous days in order to identify the preservative duration for the samples.6.Determination of the normal reference values of salivary levels of UN, Cr and UA in healthy controls:The concentrations of UN,Cr and UA in 149 healthy controls were measured in saliva and the normal reference values were determined.7.Correlation analysis of UN,Cr and UA in saliva and serum in healthy individuals:Saliva was collected in 149 healthy individuals by means of citric acid swab stimulus method,and serum samples were collected,The concentrations of UN,Cr and UA in saliva and serum were measured and the correlation of UN,Cr and UA were analysed between saliva and serum in healthy individuals.8.Correlation analysis of UN,Cr and UA in saliva and serum in patients with chronic renal failure:saliva was collected from 57 patients with chronic renal failure after garling by means of citric acid swab stimulus method and the serum samples were getted at the same time. The concentrations of UN,Cr and UA in saliva and serum were measured and the correlation of UN,Cr and UA between saliva and serum were analysed in patients with chronic renal failure.9.Determination of UN,Cr and UA in saliva and serum of patients with chronic renal failure before and after dialysis:Saliva and serum samples were collected from 19 patients with chronic renal failure before and after dialysis.The concentrations of UN,Cr and UA in saliva and serum were measured and the correlation of UN,Cr and UA between saliva and serum before and after dialysis were analysed.Results:1.The influence of different collecting method of saliva on the levels of UN,Cr and UA:the concentrations of UN,Cr and UA of saliva showed no significantly difference in two collecting methods 2.The concentrations of UN,Cr and UA of saliva collected before and after gargling was not significantly difference(P>0.05).3.The concentrations of UN,Cr and UA of saliva collected before and after lunch showed no significantly difference(P>0.05).4.The concentrations of UN,Cr and UA of saliva collected at 6 Am,7 Am,8 Am and 12 Am,as well as 1Pm and 4Pm were similar to each other and showed no significantly difference(P>0.05).5.The concentrations of UN,Cr and UA of samples stored in -20℃with 6 continuous days in healthy individuals showed no significantly difference(P>0.05).6.The concentrations of UN,Cr and UA in healthy controls fit Gaussian distribution.The reference ranges of concentrations of UN,Cr and UA were(0.93-7.33)mmol/L,(6.83-19.41)μmol/L and(30.44-418.52)μmol/L respectively with(?)±2 S method.7.The relationship of concentrations of UN,Cr and UA in saliva and serum were positively correlated in healthy individuals and the value of r were 0.566,0.169 and 0.155respectively.The UN levels in saliva and serum showed no significantly difference(P>0.05),while the levels of Cr and UA in saliva and serum were significantly different (P<0.01) and the concentrations were 13.12±3.14μmol/L vs 136.93±14.56μmol/L,224.48±99.00μmo l/L vs 296.36±81.42μmo l/L8.The concentrations of UN,Cr and UA in saliva were positively correlation with those in serum in patients with chronic renal failure and the correlation coefficient for UN,Cr and UA was 0.936,0.613 and 0.217,respectively.9.The concentration of UN,Cr and UA from patients After dialysis were significantly decreased both in saliva and serum.The decreased rates for UN,Cr and UA were 45.10%/48.63%,46.57%/46.57%and 68.47%/69.19%in saliva and in serum,respectively.Conclusions:1.The concentration of UN,Cr and UA does not influenced by collecting method for saliva.The concentration of UN,Cr and UA in saliva showed no significantly different with gargling and dining2.The collection of saliva for determination of UN,Cr and UA can be performed at any time during the day.3.The concentrations of salivary UN,Cr and UA are not affected when the saliva samples were stored in -20℃for one week.4.The concentration of salivary UN is the same with serum,while Cr and UA lower than serum.5.The concentrations of UN,Cr and UA of saliva show positively correlated with serum. 6.The difference concentrations of UN,Cr and UA in patients with chronic renal failure before or after dialysis can be detected with saliva.7.The detection of UN,Cr and UA levels in saliva were reliable and that have definite values.
