Puerarin is the main component of Radix puerarin injection solution, widely used in clinical treatment of cardiovascular disease. This experiment will establish puerarin in plasma and saliva samples HPLC determination, different doses of puerarin in rats in vivo pharmacokinetic and dynamic observation of puerarin concentration in saliva and blood in the correlation. Of puerarin using saliva for pharmacokinetic study and clinical application of therapeutic drug monitoring (TDM) the feasibility of using saliva for the injection of Chinese medicine to provide a new method to carry out TDM, a new foundation.TDM using saliva to carry out the advantage of saliva drug concentrations can be played directly reflect the pharmacological effects of free drug in the body in the process. In this study, reference chemicals in saliva pharmacokinetics of the basic theory and methods based on the pharmacokinetics of Chinese characteristics by sample handling, determination of selection conditions established applies to contain a trace amount of blood and puerarin Determination of saliva sample processing and HPLC method selectivity, accuracy, good reproducibility, high sensitivity, is able to meet the analytical requirements of this study. In particular, very low levels in saliva components, sensitivity of the method meet the salivary components measured pharmacokinetic study.Intravenous injection of high, medium and low doses of puerarin, the collection of plasma, saliva samples were measured after which the concentration of puerarin using the 3P97 pharmacokinetic software program to process data, high, medium and low-dose pharmacokinetic parameters under as follows: t1/2=13.86min,AUC=8707.93ug·min·mL-1,Vc=675.3mL,CL=0.0091mL·min-1;t1/2=11.35min,AUC=4766.73ug·min·mL-1,Vc=489.4mL,CL=0.0084mL·min-1;t1/2=12.16min,AUC=1765.09 ug·min·mL-1,Vc=500.2mL,CL=0.0133mL·min-1。High, medium and low oral dose, submandibular and parotid saliva concentrations of puerarin were: 0.154.95,0.312.36,0.150.90μg·mL-1;0.204.90,0.432.46,0.170.92μg·mL-1,High, medium, low dose, the submandibular and parotid gland of S and P correlation coefficients were: 0.9539,0.9564,0.8557,0.9479,0.92,0.8346 (P<0.05),S and P was significantly correlated. By comparing different doses of puerarin injection from the distribution of blood to the saliva, indicating that the tested components rapidly by the migration of blood to the saliva, and the factors by drugs and saliva and the salivary glands own physiological factors. To determine the concentration of saliva and blood composition in free drug concentration ratio CS CPUas an index of blood concentration of puerarin concentration correlated with the saliva analysis showed that puerarin dose range in the submandibular and parotid saliva concentration of free drug concentration in blood was significantly correlated, indicating the drug concentration in saliva can reflect the blood level of free drug concentration can be considered through the saliva instead of blood for pharmacokinetic studies of puerarin. In this study, drug concentrations for the use of saliva instead of blood drug concentration monitoring, the implementation of traditional Chinese medicine injection TDM provides a reference and research base. While traditional Chinese medicine injection widens the method of TDM study also demonstrated the clinical use of saliva for TDM feasibility of implementation.
