Objective: To observe the change of plasma ceramide and vascular endothelial dilatation function of patients with type 2 diabetes mellitus, and explore the relationship between plasma ceramide and vascular endothelial dilatation function.Methods: 41 patients with type 2 diabetes mellitus in our study, another 24 persons served as control group. Their blood pressure, height, weight, waist circumferences, hip circumferences, fasting plasma glucose, fasting serum insulin, hemoglobin Alc, triglyceride, total cholesterol, high density lipoprotein, low density lipoprotein were measured; fasting plasma ceramide was analyzed by Liquid Chromatography-Mass Spectrometry; vascular endothelial dilatation function was measured by high-resolution ultrasound. Compared plasma ceramide and vascular endothelial dilatation function of two groups by two-sample t-test, analyzed their correlation of plasma ceramide and vascular endothelial dilatation function.Results: Compared with control group, the level of plasma ceramide in type 2 diabetic patients was higher (P<0.01), the injury of vascular endothelial dilatation function (FMD,EID) in type 2 diabetic patients was more obviously (P<0.01), Pearson correlation analysis showed a negative correlation between plasma ceramide and FMD (P<0.05), Correlation Coefficient is -0.466, but no correlation was found between plasma ceramide and EID, FBS, FINS, HOMA-IR, TG, TC, HDL, LDL, BMI, WHR (P>0.05).Conclusions: The level of plasma ceramide is increased in type 2 diabetic patients, it suggested that ceramide maybe have influence on the developing of type 2 diabetes mellitus. The flow-mediated dilatation (FMD) and endothelium independent dilatation (EID) are damaged in type 2 diabetic patients. Plasma ceramide has negative correlation with the flow-mediated dilatation (FMD), it suggested that ceramide maybe have influence on the vascular endothelial dilatation function of type 2 diabetic patients.
