Objective: To explore the relationship between endotoxin and matrix metallo- proteinases-2 in the experimental liver fibrosis in rat , and the mechanism to form hepatic fibrosis related to the former.Methods: An experimental CCl4 induced hepatic fibrosis rat model was establi- shed by intraperitoneal administration of carbon tetrachloride for 2,4,6 weeks,make use of immunohistochemisty staining to semi-quantitatively determinate and observe the dynamic expression of MMP-2 andα-SMA in 2,4,6 weeks,and adopt reticular fib- er staining method to estimate the degree of liver fibrosis ,meanwhile use the ELISA method to determine the level of plasma endotoxin.Results: (1) Endotoxemia have occurred in our rat model.The plasma endotoxin levels(0, 2, 4, 6 weeks) were increased significantly, and is closely related to the expression of MMP-2 andα-SMA in the liver. (2) The expression of MMP-2 is always in accordance with theα-SMA`s in the site of expression ,time and content, especially in the central veins around, header aera, and fibrous septum,suggestiong that MMP-2 is mainly synthetized by the activated hepatic stellate cell. (3)There are a direct correlation among degree of hepatic fibrosis, MMP-2 andα-SMA expressed in liver.Conclusion: The endotoxemia is closely related to the expression of MMP-2 andα-SMA in the liver, which are closely related to the degree of hepatic fibrosis, suggestiong that MMP-2 is likely to be a important factor which the endotoxemia induces the development of hepatic fibrosis.
