Objective:To observe the changes of glycometabolism,lipid metabolism,secretary function ofβcells and the influence of apoptosis onβcells of obese rats and type-2 diabetic rats by the treatment of bezafibrateMethods:75 Sprague-Dawley rats were divided into Normal control (NC,n=10) and High fat-fed diet (HF,n=65).After the obese rats model had been formed,we divided them into two groups.One would be injected with STZ while the others wouldn't.The animal model of type 2 diabetes would be formed.Then we divided each of them into two groups again,there were normal control (NC),obese rats (OB),obese rats with bezafibrate (OB+BZ),diabetes rats (DM)and diabetes rats with bezafibrate (DM+BZ).To detect the levels of Fasting blood glucose (FBG), Triglyceride (TG), Total cholesterol (TC), Fasting insulins (FINs) et al before and after the treatment of bezafibrate.After 12 weeks intervention,dyed with Hematoxylin-Eosin, the morphous of pancrease islet were observed by microscope.The apoptoticβcells in islets were detected and quantified by the TUNEL technology.Results:1,The levels of TG,TC were decreased conspicuously in OB+BZ and DM+BZ groups.There shows lower levels of FINs in OB+BZ group than pre-intervention [(31.81±2.18)mIU/l vs (22.33±2.85)mIU/l, P<0.01],Levels of FBG in DM+BZ group were deceded than pre-intervention [(19.93±1.85)mmol/l vs (10.57±1.73)mmol/l, P<0.05],when the levels of FINs were improved[(15.84±1.73) mIU/l vs (19.50±1.64)mIU/l, P<0.05];2,The rates of apoptoticβcells in islets were higher in OB and DM groups than in NC group which conspicuously higher in DM group. After the bezafibrate treatment,the rates of apoptoticβcells in islets were decreased both in OB+BZ [(8.6±1.2)% vs (13.4±1.5)%, P<0.05] and DM+BZ [(12.6±2.1) % vs (22.6±2.5) %, P<0.05] groups.Conclusion:Long term high fat fed diet could induce obesity on the experimental rats,both the obese rats and the diabetic rats most complicated with the disorder of blood-fat.There also be observed the apoptosis ofβcells in pancrease islet of these rats.By intervening hyperlipemia with bezafibrate,it could decrease or inhibit the lipoapotosis ofβcell,then improve the secretary function ofβcell and the insulin sensitivity.So Bezafibrate may play a protective role onβcells of pancrease islet.
