| Objective: L02, HepG2, SGC7901 and SW480 cells were researched. To observe whether stem cell factor (SCF) mRNA and protein were expressed in the four cells and their levels of expression. To compare the difference of SCF content between L02 and HepG2. In oreder to explore its relationship with the occurrence of hepatoma. To study the difference of level of SCF expression in inhibition groups and non- inhibition groups of SGC7901, SW480, HepG2. In oreder to explore relationship of SCF with gastric cancer,colon carcinoma and hepatoma. To illuminate the role of SCF in the tumor of digestive system.Methods: MTT assay was used to analyze 50% concentration of inhibition (IC50). 1/2IC50 was used as concentration of experiment drugs. SCF mRNA was detected by reverse transcripation polymerase chain reaction (RT-PCR) whether it was expressed in the above four cells. SCF protein was detected by western blotting whether it was expressed in the above four cells. Logarithmic phase of L02, HepG2, SGC7901 and SW480 cells respectively culture 24h. They were randomly divided into inhibition groups and non-inhibition groups. Inhibition groups were incubated in mediuns containing CDDP for 24 hours. Non-inhibition groups were incubated in mediuns without containing CDDP for 24 hours. The relative level of SCF mRNA expression was detected by RT-PCR,and SCF protein expression was detected by western blotting. To explore relationship of SCF with tumor in digestive system.Results: IC50 of CDDP were 76.7±0.4μmol / L, 40.6±0.6μmol / L, 54.8±0.2μmol / L and 66.9±0.2μmol / L on L02, HepG2 , SGC7901 and SW480 cells, respectively. SCF mRNA and protein were expressed in the four cells. The level of SCF mRNA expression were 0.767±0.012,0.617±0.038,0.589±0.087,0.546±0.077. The level of SCF protein expression were 0.905±0.005,0.713±0.060,0.353±0.055,0.606±0.165。After four cells were inhibited by CDDP, the level of SCF mRNA expression were 0.484±0.005,0.250±0.080,0.401±0.065,0.167±0.025; The level of SCF protein expression were 0.630±0.013,0.440±0.115,0.293±0.072,0.317±0.046. Compared with non-inhibition groups, SCF expression was decreased in inhibition groups of L02 and HepG2 (P<0.05). Compared with L02 non-inhibition groups, SCF expression was decreased in HepG2 of non-inhibition groups (P<0.05). Compared with non-inhibition groups, SCF mRNA expression was decreased in inhibition groups of SGC7901,SW480 and HepG2 (P<0.05). Compared with non-inhibition groups, SCF protein expression was decreased in SW480 and HepG2 of inhibition groups (P<0.05), except SGC7901(P>0.05).Conclusions: SCF mRNA and protein were expressed in L02, HepG2, SGC7901 and SW480 cells. The level of SCF expression in L02 was higher than in HepG2. The results prove that SCF was expressed in fetal liver cell at a high level. After proliferation of L02, HepG2 cells were inhibited by CDDP, SCF mRNA and protein expression was decreased. The result indicated that SCF expression was downregulated, after hepatic tissue cancerization. It is possibility that SCF related to occurrence of hepatoma. SCF mRNA expressions were downregulated, after proliferation of SGC7901, SW480 and HepG2 cells were inhibited by CDDP. SCF protein expressions were downregulated(P<0.05), except SGC7901 . The results suggest that SCF play an important role in proliferation of colon cancer cell and hepatoma cell. We presumed that SCF expression was upregulated after stomach, colon, liver cancerization. To understand the cancerization situation, the levels of SCF mRNA and protein expression were detected in cells. Monitoring, regulation and intervention of SCF expression may be new auxiliary diagnosis or clinical treatment of colon carcinoma, hepatoma. |
