Objective Senile cataract-induced blindness is still the main causes of blindness all over the world,although surgery are increasingly sophisticated, carried out the growing popularity, after cataract also can not be completely avoided, and postoperative complications are also the impact of visual recovery. Early drug prevention and treatment of cataract is therefore of great practical significance. Oxidative damage are the main reason for the incidence of cataract. In this study, rabbit lens in vitro methods set up lens oxidative damage model, from the form, function and other aspects of propofol on oxidative damage in rabbit lens water channel protein 1 expression and its effect on the protective the lens mechanism for the clinical looking for a safe, effective prevention of cataract drugs to provide preliminary experimental evidence.Methods Rabbit's lenses cultured in vitro were divided into 3 groups: control group, oxidative stress group and groups treated with propofol (4ug/ml,40ug/ml,100ug/ml). After 24 hours of incubation, alteration of opacity was observed by photography and picture analysis; The expression of AQP1 was detected by immunohistochemistry with semiquantitative analysis. The content of the superoxide dismutase(SOD) and malonyldialdehyde(MDA) were measured by spectrophotometer. The histomorphology of lenses were examined under light microscope and the lenses ultrastructural changes were detected by electron microscopy.Results Compared with control group,xidative injury group lens opacity was shaped opaque,all the Man-like white;lens SOD decreased, MDA content increased, AQP1 expression was significantly decreased,;generally anterior capsular thickening, epithelial cell structure disorder, the surface microvilli disappeared, cytoplasm condensed, see a large number of intracytoplasmic vacuoles of various sizes, nucleocytoplasmic ratio increased two-tier structure of nuclear membrane unclear, nuclear chromatin congregate and some margination.Compared with xidative injury group, propofol group showed mild cortical lens opacities, equatorial vacuoles can be seen a small number of core basic clearly visible, lens SOD content increased, MDA decreased and AQP1 expression increased , lens ultrastructure injury mitigate.Couclusion:1. Aquaporin 1 may be involved in pathophysiologic process of lenses oxidative stress .2. It is true that some retinal ganglion cells are loss by apoptosis.3. Propofol can protect lens against oxidative stress by maintaining regulating expression of AQP-1 and production of SOD,MDA in rabbit's lenses.
