Potropaic And Therapeutic Effect Of Egg-yolk Antibody On Experimental Helicobacter Pylori Infection In BALB/C Type Mice

Objective: To establish a model of BALB/C mouce infected by HP and to evaluate the function of the inhibitory and therapeutic effect of egg-yolk antibody on experimental Helicobcter Pylori infection in BALB/C type mice with atrophic gastritis(AG).Method: To immune laying hens by killed vaccine from HP type strain (ACTT43504/NCTC11637) . Laying hens were immunized at various concentration using different adjuvants. When the titer of eggs is enough, Sterilizing the eggs 10 minites, and collecting yolk sterilely. 60 BALB/c type mice were fed one week and divided randomly into five groups. Mice in groupâ… were inoculated with sodium chloride solution; Mice in groupâ…¡,â…¢andâ…£were inoculated respectively with antibody in different doses, and then were inoculated with H.pylori; Mice in group V were inoculated with H.pylori in order to establish AG of experimental Helicobacter Pylori infection in BALB/C type mice. After 8 weeks, all the mice were sacrificed. The infection of H.pylori in gastric mucosa of the mice was analyzed by urease test and culture. The histopathological changes in the gastric mucosa of mice were assessed in H-E stained sections.Results: The optimal antigen concentration was 10~9 cfu/ml. The immunization enhancement of CpG adjuvant was stronger. There were more HP in the stomach on 8th week after inoculation and the infection rate was 100% in groupâ… andâ…¤. The infection rate was more than 70% in groupâ…¡, and less than 30% in groupâ…¢andâ…£.Conclusion: The level of specific IgY in yolks of immunized hens was affected by concentration of antigen and quality of adjuvant. The egg-yolk antibody against HP swallowed might decrease HP infection in the stomach of BALB/C mice. The protection of the antibody had intimate relationship to the dose of the antiboy.
