Objective: To study the expression of GFAP and TNF-αduring lipopolysaccharide- induced white matter damage in the early neonatal rat and the protection of xuebijing.Method: Establishment of model:20 Wistar pregnant rats were randomly divided into two groups: LPS(n=12)and saline(n=8).Pregnant rats were consecutively injected with LPS(450μg/kg)or saline on gestation d 16 and 17.The pups whose gestation age was less than 22 days were preterm pups and were removed in both groups. We selected randomly saline neonatal rat group A(n=8)and LPS neonatal rat group B(n=8),C(n=8),D(n=8).2,Early interfere: The 7 days rats(B,C)were therapy by injected with xuebijing(8ml/kg·d, 4ml/kg·d,) and the rats(A,D)were injected with saline (4ml/kg·d, 4ml/kg·d,). Neurobehavioral observation and appraising cerebral palsy: The Neurobehavior of rat was observed on 21d.The items of neurobehavioral observation included sensorimotor movement(hanging test and inclined plane test) voluntary movement, non-autokinetic movement.The data was analysed by the science of statistics.The cerebral palsy rats must have abnormal neurobehavior in aspects of sensorimotor movement and posture and muscle tension at the same time, accompanying or non-accompanying with the abnormity of non-autokinetic movement and emotional behavior,but the result of deflected plate test must be positive.4,Immunohistochemistry detection: The GFAP and TNF-αwere detected in brain sections of 21 day rat.Results: Compared with saline group,1 pregnant rat were dead after injected LPS. There were 8 pregnant rats of pre-parturition in LPS group. The rest of pregnant rats delivered 45 live mature rats and 17dead rats in LPS group. In reverse, there were no dead or pre-parturition's pregnant rats in saline control group. The pregnant rats delivered all 65 live mature rats and no dead rats in saline control group. There were a large amount of neutrophilic granulocyte in the uterus and placenta in LPS-treated group. There was obvious increase about TNF-αexpression in PWM in D group of 21 day rats, contrasted with the one in A group of 21 day rats(P<0.01);There was decrease about TNF-αexpression in B and C group of 21 day rats, contrasted with D group of 21 day rats(P<0.05),it was no obvious decrease in B and C, (P>0.01); There was obvious increase about GFAP expression in PWM in B and C group of 21 day rats, contrasted with the one in A group of 21 day rats(P<0.01);There was decrease about GFAP expression in D group of 21 day rats, contrasted with B and C group of 21 day rats(P<0.05),it was no obvious increase in B and C, (P>0.01).Conclusion:1. Chinese medicine of huayujiedu can adjust the expression of GFAP and TNF-αduring lipopolysaccharide-induced white matter damage in the early Neonatal rat 2. Chinese medicine of huayujiedu can treat intrauterine infection-induced white matter damage in the early Neonatal rat ,the mechanism may be related to removal of TNF-αand increase in GFAP expression.3. We conclude that cerebral palsy induce by intrauterine infection could treated by the Methods of huayujiedu in the theory of Traditional Chinese Medicine .
