Objective: To study the apoptosis,cytokine and B-type natriuretic peptide effected by Kangdaxin in heart failure rats, trying to explore the mechanism of Kangdaxin treat the HF from the molecular biology aspects. On the basic of western treatment perscription, plus the Kangdaxin, observed the changes of the apoptosis, cytokine and the levels of BNP after treatment in different groups.Methods: The study using the doxorubicin(Dox) induced the rat failing hearts, divided the SD rats into control group, model group, Kangdaxin group(Chinese Traditional Medicine), Irbesartan group(western medicine) and Kangdaxin plus Irbesartan group(Chinese traditional medicine plus western medicine), after four-week treatment, To observe general status, mortality rate, damaging changes in histomorphology of cardiac muscle in rats; and to measure myocardial cell apoptosis with TUNEL method, and BNP,cytokine expression in blood serum with ELISA technique.Results: 1. Compared with mortality rate in different group rats, the mortality rate in thetherapeutic groups was more remarkably lowered than in the model group. 2. Compared withmyocardial cell apoptosis in rats, there were significant difference between control group andmodel group, Compared the therapic groups with the model group has remarkable statisticalsignificance (P<0.01), Compared the Chinese traditional medicine plus western medicinegroup with the western medicine group was statistical significance (P<0.05), There were nostatistical significance between the Chinese traditional medicine group and the westernmedicine group. 3. The level of BNP,TNF-a and IL-6: The model group level of BNP,TNF-aand IL-6 was obviously higher than the control group (P<0.01);the level of BNP,TNF-a andIL-6 has decrease in groups in treatment, compared the therapeutic groups with the modelgroup was conspicuous difference; The level of BNP,TNF-a and IL-6 of the Chinesetraditional medicine plus western medicine group was lower than the western medicine group;There was no difference between the Chinese traditional medicine group and the westernmedicine group.Conclusion: The Dox successfully made the CHF animal model by deduced myocardial contractility; The Kangdaxin improved the rats in CHF general status and decreased the mortality rate. It can reduce the pathologic damage in myocardium of rats and inhibit myocardial apoptosis, the medicine can reduce the level of TNF-a, IL-6, also improve heart function; The level of BNP was the important indicatrix of the CHF diagnose and therapeutic effect. Therefore, it can prevent the progress of heart failure. The study elucidated the mechanism of Kangdaxin on treatment on the heat failure from the level of cellular, it provided the experimental base for Kangdaxin in treatment of the heart failure.
