background: Gastric carcinoma is one of the most predominant malignant tumors which threatens peoples'life in China, with the highest incidence rate in digestive tumor. Due to economic reason, many patients are late when they know that they have got this disease. Current state of gastric carcinoma is low in early diagnosis, high in incidence rate and high in mortality. There are many ways to therapy gastric carcinoma containing operation, chemotherapy, radiotherapy and biotherapy and so on. Consequently, consummating early diagnosis and improving treatments may decrease mortality or prolong life span.Objective: Detecting Helicobacter pylori(Hp) infection in groups of gastric carcinoma and normal, and approaching the relations between Hp infection and gastric carcinoma; Checking positive expression of Hp-CagA antibodies in peripheral blood in groups of gastric carcinoma and other diseases, and examining the correlation of expression of Hp-CagA antibody between peripheral blood and portal vein in group of gastric carcinoma, so to probe the action of Hp-CagA antibodies in generation and development of gastric carcinoma; Detecting expressions of WWOX protein and C-Jun protein in tissues of gastric carcinoma and normal gastric tissues, so to analyze the differences and relations in them and the relations between the expressions of them and Hp infection, moreover presume the molecular mechanism that WWOX gene and c-Jun gene induce cancerization. According to above-mentioned researches, they may offer help in prevention, cancerigenic mechanism and prognosis judgment of gastric carcinoma from diverse aspects. Methods: In the present study, we check up Hp utilizing BAMB (boric acid methylene blue staining) or RUT (rapid urease test) in 60cases of group of gastric carcinoma and 40 cases of group of superficial gastritis and normal, detecting expression of Hp-CagA antibody in peripheral blood or portal vein in group of gastric carcinoma and 40 cases of other diseases using western blotting, and examining expression of WWOX protein and c-Jun protein between tissues of gastric carcinoma and normal gastric tissues by IHC (immunohistochemisty).Results: (1) The rate of Hp infection in tissues of gastric carcinoma is higher than that in normal tissues, and Hp infection is related with infiltration and lymph nodes of tumor, all differences are significant (p<0.05); (2) The different expressions of WWOX protein between tissues of gastric carcinoma and normal gastric tissues(28.3%vs81.7%) are significant (p<0.05), and WWOX is related with cell differentiation of gastric carcinoma. The positive expressions in group of high differentiation and middle differentiation are higher than that in group of low differentiation; (3) There are different expressions of WWOX protein in group with Hp infection and group without Hp infection, but no statistical significance; (4) The positive expressions of C-Jun protein between tissues of gastric carcinoma and normal gastric tissues(68.3%vs23.3%) have significant difference(p<0.05), and it is related with infiltration and lymph nodes, the deeper tumor infiltrate, the higher expressions of C-Jun protein tumor tissues have; (5) The positive expressions of C-Jun in group with Hp infection are higher than that in group without Hp infection(78.9%vs50.0%), and it has statistical significance(p<0.05); (6) There is a difference between expressions of WWOX protein and C-Jun protein in tissues of gastric carcinoma(28.3%vs68.3%), it has statistical significance(p<0.05); (7) Positive expressions of Hp-CagA antibody of peripheral blood in group of gastric carcinoma is higher than that in group of other diseases(p<0.05), and positive expressions of Hp-CagA antibody in portal vein are higher than that in peripheral blood in gastric carcinoma, but no statistical difference.Conclusion (1) Hp infection in group of gastric carcinoma is higher than normal, it maybe lead to gastric carcinoma by inhibiting WWOX gene or activating c-Jun gene. Hp, WWOX gene and c-Jun gene possibly participate in generation of gastric carcinoma in common; (2) expressions of WWOX protein in gastric carcinoma are apparently lower than that in normal gastric tissues, it indicates that WWOX gene can prohibit cancerization as well as generation and development of gastric carcinoma; (3) C-Jun protein is related intimately with gastric carcinoma, it infers that activation of c-Jun gene maybe induce cancerization of gastric carcinoma; (4) Expressions of Hp-CagA antibody are apparently high in gastric carcinoma, and has intimate relation with gastric carcinoma, according early treatment of Hp infection, it is possible to prevent or decrease the rate of gastric carcinoma.
