Antioxidant Effects Of Vitamin A And Budesonide On The Persistent Asthma Rats

Objectives Through the establishment of the rat persistent asthma model, we took vitamin A and budesonide as the interventions, then compared the activity of SOD and level of MDA,NO in serum,lung and spleen tissues in different weeks. At the same time, we try to investigate the pathogenesis of asthma and provide the theory basis for vitamin A in the prevention and treatment of bronchial asthma.Methods 40 rats were divided into A, B, C ,D four groups randomizedly, Group A for the Control group,Group B for Asthma group, Group C for the Budesonide group, Group D for Vitamin A group. There were 10 rats in each group. On the 1st and 8th day of experiments, B, C,D groups were injected into abdominal cavity with sensitization fluid of 1.0 ml / only / times. On the 15th day of the experiments, the rats had been inhaled 1% OVA solution for 3 weeks in order to establish persistent asthma model, daily one time, 30 minutes each time. The intervention of these three groups were as follows, each group was given the 1%OVA inhalation to carry on the stimulation as usual every morning, Group C was inhaled 0.02% budesonide saline solution and vitamin A 10U/ ml for Group D every afternoon. Each group had been inhaled for 30 minutes, a total of a week. Half of rats of each group were killed on the 4th week and the 5th week after stimulation. Then the contents of SOD,MDA,NO in serum,lung and spleen tissues were measured.Results (1) After the 4th week of stimulation:The contents of SOD in serum,lung and spleen tissues of Asthma group were lower and that of MDA,NO higher than the other three groups. All the differences were significant except for the differences with SOD,MDA in spleen tissues in the vitamin A group. Compared with Control group, all contents of Vitamin A and Budesonide group were different significantly. At the same time, the differences between Vitamin A and Budesonide group were significant. (2) After the 5th week of stimulation:The contents of SOD in serum,lung and spleen tissues of Asthma group were lower and that of MDA,NO higher than the other three groups. All the differences were significant.Compared with Control group, all contents of Vitamin A and Budesonide group were different significantly, except for the difference with SOD in spleen tissues in Budesonide group. At the same time, the differences between Vitamin A and Budesonide group were significant, except for SOD in spleen,MDA in serum and lung. (3) After the 5th week of stimulation:The differences in Budesonide group were significant except for SOD in spleen tissues and NO in serum.When compared with the 5th week after stimulation, all the differences were significant in Vitamin A group.Conclusions (1) The activity of SOD were significantly decreased in in the serum, lung and also the spleen of asthma rats,while the contents of MDA and NO were significantly high.These may suggest that radicals may play a great role in the progression of asthma. (2) The inhalation of both a low dose of vitamin A and budesonide can increased the activity of SOD in the serum,lung and also the spleen of asthma rats,but on the other hand,the contents of MDA,NO were significant decreased.Such effects can last for one week after withdrawal. These may suggest that both of them have some antioxidant effects. (3) The inhalation of a low dose of vitamin A has some therapeutic effect on persistent asthma rats,and this therapeutic efficacy is weaker than that of budesonide.
