Background Psoriasis (OMIM *177900) is a common, chronic and recurrent, inflammatory skin disease, the most common characteristics of which are red, well-demarcated and heavily scaled plaques. The pathogenesis of psoriasis might be complex and unclear, involving both genetic and environmental risk factors (infection, trauma, stressful events et al). Psoriasis vulgaris (PV) is the most frequent clinical type, more than 90 percents. We found a distinguished correlation of rs3213094 polymorphism in IL12B gene with psoriasis vulgaris in Chinese Han Population by psoriasis genome-wide association study (GWAS).Objective To investigate the potential association of IL12B gene polymorphism and some phenotypes of psoriasis vulgaris in Chinese Han Population basing on stratifying analysis of the age of onset, family history and clinical types, and to provide an important genetic basis in exploring the pathogenesis of psoriasis.Methods We analyzed the distributions of genotype and allele of rs3213094 (polymorphic site within IL12B gene) in 1,139 cases and 1,132 normal controls, which derived from the psoriasis GWAS carried out by our group. The appropriate application of social science statistical software package SPSS 15.0 was used for statistical analysis of data. Chi square test was used for comparing genotypic or allelic frequencies distribution among the groups. Results 1. The genotypic frequencies of rs3213094 (polymorphic site within IL12B gene) were of statistical significance between the cases and the controls (χ2=22.50, df=2, P=1.30×10-5), there was a similar trend for the allelic frequencies (χ2=21.18, df=1, P=4.2×10-6, OR 1.32, 95%CI 1.17-1.49). 2. The allelic frequencies of rs3213094 were statistical significance between the pediatric age of onset cases and controls (χ2=5.55, df=1, P=0.02, OR 1.23, 95%CI 1.03-1.47), The genotypic frequencies were not (χ2=5.59, P=0.06). The genotypic and allelic frequencies of rs3213094 were also statistical significance between the adult age of onset cases and controls (genotypeχ2=23.17, P=9.32×10-6; alleleχ2=21.18, df=1, P=4.2×10-6, OR 1.36, 95%CI 1.36-1.55). However, there were no statistical significance between the pediatric age of onset and adult age of onset cases (genotypeχ2=2.29, P=0.32, PAA=0.13, PAG=0.53, PGG=0.68; alleleχ2=1.08, df=1, P=0.30, OR 0.91, 95%CI 0.75-1.10). 3. The allelic frequencies of rs3213094 were statistical significance between the familial cases and controls (χ2=4.37, df=1, P=0.04, OR 1.19, 95%CI 1.01-1.39), The genotypic frequencies were not (χ2=5.67, P=0.06). The genotypic and allelic frequencies of rs3213094 were also statistical significance between the sporadic cases and controls (genotypeχ2=23.65, P=7.32×10-6; alleleχ2=23.32, df=1, P=1.40×10-6, OR 1.40, 95%CI 1.22-1.61). However, there were no statistical significance between the familial and sporadic cases (genotypeχ2=4.04, P=0.13, PAA=0.33, PAG=0.22, PGG=0.05; alleleχ2=3.52, df=1, P=0.06, OR 0.85, 95%CI 0.71-1.01). 4. The genotypic and allelic frequencies of rs3213094 were statistical significance between the acute guttate cases and controls (genotypeχ2=11.09, P=0.004; alleleχ2=10.86, df=1, P=9.85×10-4, OR 1.33, 95%CI 1.12-1.57). The genotypic and allelic frequencies of rs3213094 were also statistical significance between the chronic plaque cases and controls (genotypeχ2=17.57, P =1.52×10-4; alleleχ2=16.59, df=1, P=4.64×10-5, OR 1.32, 95%CI 1.15-1.51). However, there were no statistical significance between chronic plaque cases and the acute guttate cases (genotypeχ2=0.03, P=0.98, PAA=0.95, PAG=0.85, PGG=0.88; alleleχ2=0.00, df=1, P= 0.94, OR 0.99, 95%CI 0.83-1.19).Conclusions 1. The polymorphism of IL12B gene (rs3213094) is associated with the susceptibility of psoriasis vulgaris in the Chinese Han population; 2. The polymorphism of IL12B gene (rs3213094) may be not associated with age of onset of psoriasis vulgaris; 3. The polymorphism of IL12B gene (rs3213094) is not associated with family history of psoriasis vulgaris; 4. The polymorphism of IL12B gene (rs3213094) is not associated with clinical types of psoriasis vulgaris; 5. Our findings indicate that the polymorphism of IL12B gene (rs3213094) may have no effection on the age of onset, family history and clinical types, which may be associated with different psoriasis genetic basis and pathogenesis.
