| Transgenic technology has a tremendous role in promoting the development of modern biology and medicine.Transgenic mouse technology has become a conventional laboratory techniques, now thousands of different types of transgenic mice has been established which provides an effective tool in vivo for biomedical research.However, the study of transgenic rat in China is a slow progress, the transgenic efficiency is low, in order to enhance the efficiency of transgenic rats, we study the number of embryos after superovulation, embryo quality, the survival of embryo after nuclear injection and the success rate of embryo transfer of different combinations of PMSG and HCG to different week-old rats, we build a successful nuclear injection transgenic rat technology by the building of three different gene transgenic rat model.On this basis we injected the pBC-SV40Tag into the prokaryotic of SD rat embryos, the testing of PCR and Southern provide that we obtained positive SV40Tag transgenic rats. RT-PCR and westhern bloting analysis showed that: pBC- SV40Tag transgenic rats, not only express SV40Tag in pancreatic tissue as expected, but also highly expressed in the brain and we successfully obtained brain tumor transgenic rat model. |
