Influence Of Bao'shen Recipe On Expressions Of TGF-β1,VEGF And MMP-9 In Kidney Tissue In Experimental Diabetic Nephropathy Rats

Objective:To investigate the influence of Bao'shen recipe on expressions of TGF-β1,VEGF and MMP-9 in the kidney of diabetic rats.Methods:Streptozotocin(STZ) was injected through abdominal cavity to make models of diabetic rats.Wistar rats were randomized divided into normal control rat group(group C),model control group(group D) and Bao'shen recipe curative group(group DB).After administration for 8 weeks, the rats were observed indexes of blood glucose(BG),body weight(BW), kidney weight/body weight(KW/BW),blood urea nitrogen(BUN), chol(CHO),creatinine clearance rate(Ccr),urine albumin excretion rate (UAER) and pathological changes of renal tissue.Expressions of TGF-β1,VEGF and MMP-9 in renal tissue were detected by immunohistochemistry.Results:There was a significant difference in levels of the above parameters between group C and group D(P＜0.01).Compared with group D,the index of KW/BW was lower(P＜0.05),the index of Ccr was higher(P＜0.01),and the indexes of BUN,CHO,UAER were markedly lower(P＜0.01) in group DB.Compared with group D,the damage of kidney pathology mitigated in group DB.Compared with group C,the expressions of TGF-β1 and VEGF were higher,while the expression of MMP-9 was lower in renal tissue of group D(P＜0.01).The expressions of TGF-β1 and VEGF were reduced(P＜0.05),while the expression of MMP-9 was advanced(P＜0.01) by using Bao'shen recipe.Conclusion:Bao'shen recipe may have a protective effect on renal function,and alleviate the damage of kidney pathology of DM rats.The mechanism may be related to that Bao'shen recipe can depress the expressions of TGF-β1 and VEGF,and increase the expression of MMP-9 in the kidney of DM rats,as a result,it can decrease the synthesis and secretion of extracellular matrix.
