The Study Of The Glucocorticoids On The Animal Model Disc Autoimmune Effect

Objective Establishing disc autoimmune inflammue animal model by destroying the fibrous ring and the cartilage lamina through the surgery to expose nucleus pulposus,Glucocorticoids interfere with the autoimmune process,discussing the effects of glucocorticoids on the disc auto-immune inflammatory state,to provide a new hypothesis for clinical treatment.Methods Establishing the rabbit model of autoimmune by using the anterior external surgery to destroy the fibrous ring and the cartilage lamina and with 30 healthy adult New Zealand white rabbits no matter the sexual,model were randomly divided into the treatment group(15) and the control group(15).The treatment group were injected with Glucocorticoids afeter the surgery,the dose was 0.1 mg/kg. The two group were taken MR on 1 week,2 weeks,4 weeks after surgery,to be executed and get the intervertebral disc in the surgical area after took the MRI.The specimen were used to Conventional HE stain and Immunohistochemical stain; Observing the lymphocyte invasion and expression of the CD4~+ and CD8~+ lymphocyte.Discussing the effects of glucocorticoids on the disc auto-immune inflammatory state by using statistics to Analysis two groups date indicators.Result 1.Imaging findings:On 1 and 2 weeks after surgery,abnormalities on MR imaging can not be see in the two groups;On 4 weeks after surgery,the intervertebral space showed narrow,abnormal signal on intervertebral disc and low signal change of nucleus pulposus,Treatment group and control group no significant difference.2.HE stain:Under the light microscope,interior/external layer fibrous ring showed confused and lost the normal structure in two group, collagenous fibers are not continuous and hyperplasia,Fibroblasts increased.On 1 and 4 weeks after surgery,lymphocytic infiltration can be seen in the edge of the annulus fibrosus,On 3 weeks after surgery,Foreign body granuloma can be seen in the edge of the annulus fibrosus,lymphocyte infiltration can be seen in the nudeoplasty,Lymphocytic infiltration in the treatment group were reduced.3. Immunohistochemistry stain:on 1 weeks,2 weeks and 4 week after the surgery,the CD4~+ and CD8~+ lymphocyte can be seen in the area next to the surgical,The CD4~+ lymphocyte mainly expressed on 1 week,2 weeks,the CD8~+ lymphocyte mainly expressed on 2 weeks,4weeks.CD4~+,CD8~+ lymphocyte count of the treatment group was lower than the control group,two groups has statistical significance.Conclusions 1.Autoimmune inflammation animal model can be established by destroying the fibrous ring and the cartilage lamina through the surgery.2. Glucocorticoids can affect the disc autoimmune progress,to provide a theoretical foundation.
