Objective:1.To investigate the expression of Ang II and its receptor 1,VEGF,CD34 in normal ovarian tissues and ovarian tumor tissues,and analyze the correlations of the above indexes expression with several clinicopathologic characteristics.2.To detect the mRNA expression of AT1R in ovarian cancer cell lines SKOV3 and ES2.3.To detect the mRNA expression of AT1R in normal ovarian surface epithelium and ovarian tumor tissues,and analyze the correlation with several clinicopathologic characteristics.Method:1.Expression of Ang II and its receptor 1,VEGF,CD34 were detected in 20 normal tissues,20 benignin tissues,20 bordline tissues and 60 ovarian cancer patients by using immunohistochemistry. .2.RT-PCR was used to detect the mRNA expression of AT1R in ovarian cancer cell lines SKOV3 and ES2.3.RT-PCR was used to measure the mRNA expression of AT1R in 6 normal ovarian surface epithelium and 30 ovarian cancer patients.Result:1.AngⅡwas not expressed on the surface epithelium of the normal ovary,The positive expression of AngⅡwas detected in benignin tissues,bordline tissues and epithelial ovarian cancer patients,and was localized both on themembrane and in the cytoplasm of tumor cells.The positive rate of AngⅡin benignin tissues,bordline tissues and epithelial ovarian cancer patients were 25%,60%,80%respectively.The expression of AngⅡwas significantly up-regulated in epithelial ovarian cancer compared to bordline tissues and benignin tissues (p<0.05);so did bordline tissues compared to benignin tissues(p<0.05).2..AT1R was not expressed on the surface epithelium of the normal ovary,The positive expression of AT1R was detected in benignin tissues,bordline tissues and epithelial ovarian cancer patients,and was localized both on themembrane and in the cytoplasm of tumor cells.The positive rate of AT1R in benignin tissues, bordline tissues and epithelial ovarian cancer patients were 25%,60%,80% respectively.The expression of AT1Rwas significantly up-regulated in epithelial ovarian cancer compared to bordline tissues and benignin tissues(p<0.05);so did bordline tissues compared to benignin tissues(p<0.05).3.VEGF was expressed in normal ovary,benignin tissues,bordline tissues and epithelial ovarian cancer patients,and was localized in the cytoplasm of tumor cells.The positive rate of VEGF in normal ovary,benignin tissues,bordline tissues and epithelial ovarian cancer patients were 5%,15%,25%,43.33%respectively.The expression of VEGF was significantly up-regulated in epithelial ovarian cancer compared to normal ovary,bordline tissues and benignin tissues(p<0.05);so did bordline tissues compared to normal ovary and benignin tissues(p<0.05).in contrast,The expression of VEGF was not significantly up-regulated in benignin tissues compared to normal ovary(p>0.05).4.CD34 was expressed in normal ovary,benignin tissues,bordline tissues and epithelial ovarian cancer patients,and was localized in the cytoplasm of vascular endothelial cells.The MVD in normal ovary,benignin tissues,bordline tissues and epithelial ovarian cancer patients were 7.38±0.86,8.43±1.05,10.57±1.24,14.88±1.99 respectively.The MVD was significantly up-regulated in epithelial ovarian cancer compared to normal ovary,bordline tissues and benignin tissues(p<0.05);so did bordline tissues compared to normal ovary (p<0.05).The MVD was not significantly up-regulated in bordline tissues compared to normal ovary and benignin tissues(p>0.05).5.In epithelial ovarian cancer,the expression of AngⅡ,AT1R,VEGF and the MVD were not related to the age,histological type,FIGO stage,and so on(p>0.05). the expression of AT1R,VEGF and the MVD were related to differentiation (p<0.05).6.In epithelial ovarian cancer,the expression of AngⅡwas positively related to AT1R,VEGF(P<0.01)and so did the MVD(P<0.05);the expression of AT1R was positively related to VEGF and the MVD(P<0.05);the expression of VEGF was positively related to the MVD(P<0.05). 7.The prognosis of epithelial ovarian cancer patients who had negative,weakly positive expression for Ang IK AT1R,VEGF was much more better than that who had strongly positive expression;The prognosis of patients whose MVD...
