Effect Of Ganxinning Oral Liguid On CGRP And ANP In Rats With Acute Myocardial Ischemia

Objective:By observing the effect of Ganxinning Oral Liguid on electro- cardiogram (ECG)in surface,calcitonin genen-related peptide(CGRP)in blood plasma and atrial natriuretic polypeptide(ANP)in myocardial tissue of acute myocardial ischemia rats,to explore the mechanism of prevention and treatment of coronary heart disease(CHD),and to provide objective and reliable exper- imental basis for guidance clinical medicine use.Methods:60 SD rats were divided into 6 groups at random: normal control group,model control group,western medicine group(diltiazem hydrochloride,8.1mg/kg ),Ganxinning high dosage group(18.0g/kg),Ganxinning middle dosage group (9.0g/kg),Ganxinning low dosage group(4.5g/kg).Intragastric admin-istration was adopted once a day for 14 days. Acute myocardial ischemia model rat was established by injecting pituitrin from sublingual vein 1 hours after the last admini-stration.RecordedⅡlead ECG for five minutes and observed the chan- ges of high degree about J point,selected six time points:the 0 second,the 15 second,the 30 second,the 1 minute,the 3 minute,the 5 minute, measured five waveforms of each time point and take the average,recorded and calculated the changes of high degree about J point (either increased or decreased,taked the absolute value of changes).after three hours, taked 2 ml blood from the heart and detected the concentrations of CGRP, taked atrial tissue homogenate to detect the concentrations of ANP.Results: Each of Ganxinning dosage groups can significantly counteracted the ECG changes of the acute myocardial ischemia rats caused by injection of pituitrin, compared with the model group, were significant (P﹤0.05 or P﹤0.01).Compared with model group,each of Ganxinning dosage groups could significantly increased the activity of CGRP and reduced the activity of ANP(P﹤0.05 or P﹤0.01).Compared with the western medicine group,neither Guan- xinning high dosage group nor middle dosage group was significant(P﹥0.05).Conclusion:Guanxinning oral liquid can improve J point ECG changes of the acute myocardial ischemia rats,and can effectively increase plasma CGRP con- tent,reduce myocardial ANP content,the evaluation of treatment effective- ness: Guanxinning high dosage group﹥Guanxinning middle dosage group﹥Guan- xinning low dosage group. Speculate about the possible mechanism:regulating vasoactive substances,improving the endothelium,and enhancing the heart function, which plays a good role in prevention and treatment of coronary heart disease angina pectoris.
