Sea urchin belongs to phylum Echinodermata,class Echinoidea,it is the ancient organsims in the oceans. Sea urchin has many medicinal values and usually used as a food in people's normal life.In clinical we usually used shell and spine of sea urchin for curing canker,paronychia,otitis media,crewels,etc.After modern pharmacological researches,it has the pharmacological activities of toxicity,paralysis,hemolysis, hypertenst and many other potential functions.In the resent study,we found that the main chemical compositions in the Sea Urchin were pigment,toxin,Sterol,fatty acid,amino acids,vitamin,hormone,etc. Moreover,there were many inorganic salts in the animal,such as Ca,Mg,Fe,K,Na,etc.Using the conventional column chromatography methods,we carried out systemic chemical compositions on Tripneustes gratilla.The 0.8 kg dry powder Tripneustes gratilla were soaked with 85%ethyl alcohol for three times,7 days each time.The total water solution was extracts with the same volume chloroform,the water fraction was then extracted with n-butanol and the chloroform fraction was extracted with water,ethanol and n-hexane respectively,then we got the water,ethanol,n-hexane and n-butanol fractions.We isolated 17 compounds by Gel column and sephadex LH-20 chromatography method and identified 7 compounds by modern chromatography technology(MS,1H-NMR,13C-NMR) from the 4 fractions,the 7 compounds were all firstly isolated from Tripneustes gratilla.They were echolest-6-ene-5α, 8α-epidioxy-3β-itol,β-indole,dibutyl phthalat,squalus glycol,2,3-dihydroxypropyl elaidate,phenylacetic acid,cholesterin.In order to look for the potential anti-tumor compounds from Tripneustes gratilla,we screening the anti-tumor activity of the isolated compounds by cell culture in vitro.We used HepG2 uterus cancer cell, SGC-7901 stomach cancer cell,HepG2 liver cancer cell and a normal cell strain,to screening the anti-tumor activity of echolest-6-ene-5α,8α-epidioxy-3β-itol by MTT assay.We found that echolest-6-ene-5α,8α-epidioxy-3β-itol can potential inhibit the cell proliferation of the four cell strain,the IC50 values were 215μg/mL on L02 person embryo liver cell,94μg/mL on HepG2 uterus cancer cell,64.7μg/mL on SGC-7901 stomach cancer cell and 99μg/mL on HepG2 liver cancer cell respectively。We can concluded that echolest-6-ene-5α,8α-epidioxy-3β-itol can potential inhibit the cell proliferation of the four cell strain and would be a potential anti-tumor medicine after more researches.
