Diabetes is a kind of serious disease.What cause diabetes is not very clear,and there are still more work to do in this field.What's more,healthy animal and human being have different reactions on pesticide. There is little research on effects of poison on organism infected by diabetes.So this paper studied toxic effects of anticholinesterases on induced diabetic mice in order to set limit to using pesticide.And illustrate the mechanism of diabefes.Throug streptozocin- induced diabetic mice,some iadex were studies,which are pancreases the tissue slices,activity of Na~+,K~+-ATPase,effects of DDVP,Propoxur,Donepezil·HCl, Methamidophos on AChE and effects ofdonepezil·HCl on ACHE.The results are as follow:1.Making model of diabetic mice:streptozocin(100mg/kg.d)were give to seven old ICR female mices. The value of blood sugar is 300mg/dl.There are significant different(P<0.05)between diabetes group and no- diabetes group in many aspects such as drinking,body weight and emiction.2.Observing pancreases the tissue slices:observingβ-cell obtained from diabetes group and nodiabetes group.The diabetes group shows that the structure ofβ-cell is not compact,the shape abnormal, and the number decreasing compare with no- diabetes group,illuslrating that the model of diabetic mice is established.3.Determining activity of Na~+,K~+-ATPase:The index of striatum,hippocampus,cortex and Na~+,K~+-ATPase were determined.The result shows that the activity in diabetes growp decrease very much with no-diabetes group(p<0.01).4.Effects of DDVP on AChE of streptozocin-indaced diabetes mices:the activity of striatum hippocampus cortex AChE were determined after DDVP(5mg/kg) wa given to mites.There indexes are net significant difference compared with no-diabetic group.The degree of tremor and salivation is more thin in diabetes group than in no-diabetes group.5.Effect of propoxur on AChE of strepfozoocin-induced diabetes mices:the activity of AChE in stiatum,hippocampus and cortex was determined during 180 min after propoxur(10mg/kg) was give to mices,the result is as same as test four above.6.Effect os donepezil·HCl under condition of intrvion on AChE:the activity of AChE(57.98%) was limited bu donepezil(10nM),and ACHE(92.44%) was limited by donepezil(1000nM).This result show that donepezil·HCl has strong limit to ACHE.7.Effect of donepezil·HCl on AChE of streptozocin-induced diatebets mices:theresult is exactly as same as test 4 and 5 above.The activity of AChE was determined after donepezil·HCl(5mg/kg) was given to mices.8.Effects of methamidophos on AChE of streptozocin-induced diabetes mices:The activity of AChE was determined after Methamidopgos(7 mg/kg) was given to mices.The activity in diabetes group has significant difference compared with no-diabetes group(p<0.01)during 3 hours and 24 hours.Contrasted with no-diaberes group,arisen time of tremor is late and the degree of tremor and salivation is late and the degree of tremor and salivation is thin.
