ObjectiveProteolysis-inducing factor(PIF) is a sulfated glycoprotein of Mr 24,000,inducing protein degradation in skeletal muscle directly in vitro.PIF was shown to be present in the urine of patients whose rate of weight loss exceeded 1kg·month-1,who had a varied range of carcinomas,particularly pancreas,live,breast,ovary,lung,colon and rectum.The study showed PIF was secreted by mmour cell,and excreted through urine, but the relation between PIF and weight loss in cancer patients is in dispute.In the study,the purpose is to study the relation between PIF expression in nonsmall-cell lung cancer tissue and weight loss or clinical and pathological traits.And with the sensitivity comprising with CEA and CYFRA21-1,to research for its possibility as tumor marker.Material and Methods1.case source and methodPIF expression was tested in the tumor tissue of 87 patients with nonsmall-cell lung cancer by immunohistochemistry,and approach the relation between PIF expression in nonsmall-cell lung cancer tissue and weight loss or clinical and pathological traits. Then,we analysed the possibility of PIF as tumor marker by comparing with the CEA and CYFRA21-1.2.Standard of consequenceThe percentage of stained cell were taken as a measure of PIF expression.Tissue was considerd positive for PIF expression if tissue staining was found in the cytoplasm of more than 30%cells at a magnification of 400×The computed value of weight loss more than 5%was as positive.The tumor marker being more than the upper line of the normal extent is positive.CEA is more than 5.0ng/ml,and CYFRA21-1 is more than 3.3ng/ml.3.Statistics analysis It was used to analyse the data with Chi-square test and Spearman correlation.SAS 8.2 is used to analyse the data.And the significance level(a) is 0.05.A P-value of<0.05 was considered statistically significant.Result1.Expression of PIF in nonsmall cell lung cancer tissue.The experiment demonstrated that the tumor tissue of nonsmall-cell lung cancer expressed PIF,positive ratio 51.7%(45/87),the staining was mainly observed in the cytoplasm of tumour cells as brown grains.And the difference comprising with control was statistically significant(P<0.01).2.The relation between PIFexpression and weight lossThe rate of weight loss in PIF positive patients was higher than the negative ones(P<0.05).It is positive correlation between PIF expression and weight loss(r=0.25, P=0.019).3.The relationship in the expression of PIF with nonsmall-cell lung cancer clinical and pathological traits.Expression of PIF between lung adenocarcinoma and squamous carcinoma had no statistic signaficance(P>0.05),and no statistic signaficance in difference in nonsmall-cell lung cancer clinical stage(P>0.05),also no statistic signaficance in different tissue differentiation of NSCLC(P>0.05).4.Sensitivity of PIF with CEA and CYFRA21-1As tumor marker,its sensitivity is higher than CEA(P<0.05).And the sensitivity of PIF in nonsmall-cell lung cancer comparing with CYFRA21-1 had no statistic signaficance(P>0.05)Conclusion1.PIF expression in NSCLC case and the control was signaficantly difference.2.Patients with PIF positive expression had high rate in weight loss,and PIF positive expression and patients' weight loss was positive correlation.3.Its expression had no relation with tissue type,clinical stage and tissue differentiation4.It is possible for PIF as tumour marker in NSCLC.
