ObjectiveTo observe the effects of hyperbaric oxygen on the apoptosis in intestinal mucosa during the different periods of ischemia-reperfusion injury and elucidate its underlying mechanisms.MethodRats were submitted to 1 hour ischemia by clamping superior mesenteric artery, and 1 hour reperfusion by removing clamp.40 healthy male SD rats,weighing 250-300 grams,completely randomly divided into 4 groups.(1) Ischemia-reperfusion group(I/R): Anesthesia planing exposed abdominal superior mesenteric artery,clamping cause ischemic 1hour,then unclamping to reperfuse 1hour;(2) Hyperbaric oxygen preconditioning(HBO-P):After pre-giving 1 hour hyperbaric oxygen,then I/R 1 hour. (3)Ischemia group hyperbaric oxygen(HBO-I):Anesthesia planed one-hour period of abdominal ischemia simultaneously give hyperbaric oxygen,then reperfuse 1 hour;(4) Reperfusion period of hyperbaric oxygen group(HBO-R);Anesthesia planing abdominal ischemia 1 hour,1-hour period of reperfusion to hyperbaric oxygen,;using immunohistochemical detection of rat small intestine tissue aspartate-specific cysteine protease(Caspase-3) expression and observe rat intestinal epithelial cells morphological changes by light microscopy and use colorimetric detection of ATP content in intestinal tissue homogenates.Results 1,The contrast of rat intestinal tissue the content of caspase-3From HBO-I group,HBO-P group,HBO-R group,to I/R group rats small intestine caspase-3 optical density was showing an increasing trend,and HBO-I group contrast to HBO-P group,HBO-R group and I/R group,the caspase-3 optical density decreased significantly,its inter-group differences have statistical significance(P<0.05).HBO-P group contrast to HBO-R group and I/R group,the caspase-3 optical density decreased significantly,its inter-group differences have statistical significance(P<0.05).HBO-R group contrast to I/R group although it also reduced,its inter-group differences did not have statistical significance.inter-group differences did not have statistical significance(P>0.05).2,The contrast of rat small intestinal tissue homogenate content of the ATP enzymeFrom I/R group to HBO-R group,HBO-P group to HBO-I group,the content of the ATP enzyme was showing an increasing trend,HBO-I group compare to HBO-P group,HBO-R group and I/R group,the content of the ATP enzyme increased significantly,its inter-group differences have statistical significance(P<0.05).HBO-P group compare to HBO-R group and I/R group,the content of the ATP enzyme increased significantly,its inter-group differences have statistical significance(P<0.05). Although HBO-R group compare to I/R group also increased,its inter-group differences not statistically significant(P>0.05).ConclusionThere was a connection between HBO,small intestinal I/R injury,and small intestinal mucosa epithelial cells apoptosis.And the treatment with HBO can maintain energy metabolism of epithelial cells and reduce deletion of ATP,and to lighten I/R injury apoptosis of epithelial cells.In ischemic period and pre-ischemic period,the treatment with HBO showing a benefit result.The best result was obtained when HBO was administered during ischemia period,and no effect when HBO treatment during reperfusion period.
