BackgroundCervical cancer is a common female malignancy,the incidence in recent years,is a clear trend of younger,the cause has not yet been made fully aware of the current, known that high-risk human papillomavirus(HPV) infection is the major risk factors for cervical cancer,other factors probably also involved in the incidence of cervical cancer and development,which includes changes in ion channels.Calcium channel is the extracellular calcium influx of cells through the ion channel,and it exists in various body tissues and cells involved in various physiological functions of the body.A large number of studies show that calcium channel are related with tumor occurrence and development.L-type calcium ion channel(Cav1.2) is one of voltage-dependent calcium ion channels subtypes which is widely distributed in the body's normal tissue.Have been reported that Cav1.2 in the breast tumor,colon tumor,endocrine tumor and pancreatic cancer tissues,but not expressed in cervical cancer cases reported yet.The purposeofthis study is through the analysis of Cav1.2 in normal tissues of cervix,cervica -1 squamous cell carcinoma find their relation with cervical cancer.PurposeTo study the expression and clinical significance of Cav1.2α1C mRNA in cervical cancer.MethodsTotal RNA samples used in this study were isolated from 50 patients with invasive cervical cancer who underwent biopsy or curative resection,and another 10 cervical cancer specimens from the same patients who after radiochemotherapy two weeks at the First Affiliated Hospital of China Medical University.Besides,we collected 10 cases of normal cervical tissue.These tissue samples were obtained after hysterectomy because of benign uterine myoma. Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of Cav1.2α1C mRNA in 50 patients with cervical cancer,10 patients with cervical cancer after radiochemotherapy,and 10 patients with normal cervical tissue. Total RNA extraction,cDNA and subsequent PCR amplifications according to the RNA PCR Kit(AMV)Ver3.0 instruction.PCR products(5μl) were electrophoresed with 2%agarose-1×TAE gels containing 0.5μg/ml ethidium bromide and scale gray level for data analysis and statistics.Paired t-tests or nonparametric tests were used to evaluate statistical significance.Graphs and further analysis were generated with SigmaPlot5 software(Aurora,CO).Differences were considered as significance when P values were less than 0.05(P<0.05).Results1,The expression of Cav1.2α1C mRNA in normal cervix and cervical carcinomaThe expression of Cav1.2α1C mRNA were significant different between the normal cervical tissues and cervical cancers(P=0.000).2,Cav1.2α1C mRNA at different stages of cervical squamous cell carcinomaThe expression of Cav1.2α1C mRNA at different stages of cervical squamous cell carcinoma was significantly higher than normal cervical tissue.There were significant differences(P=0.000,P=0.000,P=0.001);ⅠandⅡstage cervical cancer compared,the difference has statistical significance(P=0.038);Ⅰperiod of cervical cancer andⅢ,Ⅳperiod of cervical cancer compared with the difference significant(P=0.004);Ⅱperiod of cervical cancer andⅢ,Ⅳstages of cervical cancer compared with a statistically significant difference(P=0.008).3,Cav1.2α1C mRNA in different differentiation of cervical squamous cell carcinoma differentiationCav1.2α1C mRNA inwell-differentiated,moderately differentiated,poorly differentiated cervical squamous cell carcinoma was significantly higher than normal cervical tissue,there is significant difference(P=0.008,P=0.002,P=0.009);At high, medium differentiation,compared with poorly differentiated,the difference was significant(P=0.038).4,Cav1.2α1C mRNA was higher before radiotherapy and chemotherapy than that after radio-chemotherapy,they have significant differences(P=0.003).Cav1.2α1C mRNA in cervical squamous cell carcinoma after radiotherapy and chemotherapy tissues compare with the control group,there is no significant difference(P=0.64).5,Group of normal cervix and cervical cancer group the average age of comparison,the difference was not significant(P>0.05).Conclusion1,Cav1.2 participates in the proliferation of cervical cancer cell,and plays an important role in tumorgenesis of cervical cancer.2,Cav1.2 has a positive correlation with clinical stages and pathological grades of cervical cancer.3,Radio-chemotherapy inhibits the expression of Cav1.2 in cervical cancer.
