Experimental Study On Antiaging By The Therapy Of Xing Pi Gong Tiao(XPGT)

Objective the spleen is the origin of the acquired constitution,the source of qi blood Shenghua,spleen deficiency is one of the important mechanisms of aging,Bupi method is one of the main anti-aging methods.Based on theories of "Huang Di Nei Jing" documented "The heart promotes blood,the blood promotes spleen" and "Nan Jing" documented" Tonifying the mother-organ while treating a case of deficient",this paper starts as characteristics of heart, spleen Zangxiang and their relationship,Studies comparatively anti-aging from the heart,anti-aging from the spleen,and anti-aging from conditioning the heart and spleen,focuses on the important role of anti-aging on Xin Pi Gong Tiao method.The paper establishes mainly Tiaoxin method,Tiaopi method,and Xin Pi Gong Tiao method to respectively demonstrate functions to anti-aging, in order to reveal initially the important role,the scientific connotation,the Xinpi-related possible material basis of Xin Pi Gong Tiao on anti-aging and its significance.Method 1.This experiment optional 3-month-old Kunming mice 60(purchased in the prevention of Medicine Institute of Science and Technology of Hubei Province Experimental Animal Center,Permit No:NO:0001498),were equally divided between male and female, weight 20±2 grams,a week of normal feeding in order to adapt to environment,no abnormal reaction observed,were randomly divided into 5 groups:the normal group,model group,Tiaoxin group, Tiaopi group and Xin PI Gong Tiao group respectively.Eyerygroups were 12,sub-cage rearing.Then the model group,Tiaoxin group, Tiaopi group,Xin PI Gong Tiao group were transferred in mice by subcutaneous injection every day back and neck at 10%concentration of D-galactose,injection capacity of 0.01ml·g^-1(per gram of body weight 0.01ml),once a day for 6 weeks,body weight measured once a week,depending on the capacity of the last injection weight. Resulted in subacute aging model.Normal mice carotid back daily subcutaneous injection of normal saline 0.1ml,continuous injection for 6 weeks.Subcutaneous injection in the neck back to the D-galactose model drugs at the same time,Tiaoxin group,Tiaopi group,Xin PI Gong Tiao group were transferred 0.2ml/20 grams of body weight according to the capacity of oral administration,the normal group and model group are given,such as capacity(0.2ml) of distilled water and administered in accordance with the direct calculation of equivalent dose,clinical adult dosage,the amount converted into mice,administration of Tiaoxin group of 5g/Kg/d, group administration Tiaopi of 5g/Kg/d,Xin PI Gong Tiao group were transferred to 5g/Kg/d,for six weeks medication.All mice were free diet.2.drugs:drug Tiaoxin by:ginseng,Chinese angelica, leech,Tubiechong,and Dilong,Guizhi composition containing condensed into the side 0.5g/ml Pharmacognosy of Chinese medicine preparations,according to rat body weight were converted to mice drug dose of 0.2ml / 20g / day;drug Tiaopi by:ginseng,Atractylodes macrocephala,Fu baicalensis,Zhigancao,Jiao Sanxian,Chenpi, Poncirus trifoliata,Sparganium,Ezhu composition,the parties containing condensed into pharmacognostic 0.5g/ml Chinese medicine preparations,according to rat body weight were converted to mice dose 0.2ml / 20g /day;drug Xin PI Gong Tiao by:ginseng,Chinese angelica,leech,Tubiechong,and Dilong,Guizhi,Jiao Sanxian, Chenpi,Hovenia Shell,Trigonobalanus,Ezhu composition,condensed into the side with the Chinese crude drug 0.5g/ml agents,according to rat body weight were dose conversion mice 0.2ml / 20g / day.3.six weeks after delivery were used to detect drug Tiaoxin, Tiaopi and drug Xin PI Gong Tiao were transferred to the aging mice heart,spleen SOD activity,MDA content and ultrastructure of the spleen.Results 1.Model group of mice heart,spleen SOD activity was significantly lower than the normal group,heart,spleen MDA content was significantly higher than the normal group,the ultrastructure of the spleen also occurred in the more serious losses.2.Tiaoxin medicine,Tiaopi drugs,Xin PI Gong Tiao drugs were transferred to the aging mice heart,spleen SOD activity than the model group improved,MDA content than the model group decreased significantly,in which the effect of Xin PI Gong Tiao group were significantly better adjusted in Tiaoxin group,Tiaopi group.3.Tiaoxingroup,Tiaopi group and Xin PI Gong Tiao group of aging mice transferred spleen pathological changes in the structure of a high degree of improvement,but the Xin PI Gong Tiao group was significantly superior to other groups.Conclusion The Model group's SOD activity of heart and spleen tissues increase and the MDA quantity of heart and spleen tissues decrease obviously than normal group,prove succeed in duplicate old and feeble animal model already,the therapy of Xing Pi Gong Tiao(XPGT)were transferred to effectively improve the aging mice heart,spleen SOD activity and reduced MDA content in the spleen of aging mice of the pathological changes in the structure of a high degree of improvement,to achieve the purpose of the anti-aging.
