Study on early intervention of Hartley guinea pigs intervertebral disc degeneration affected by Du huo Ji sheng DecoctionBackgroundThe cervical and lumbar spine disease which caused by disc degeneration (Intervertebral disc degeneration,IVDD) increasingly threats to people's health. Intervertebral disc degeneration is a long and not reversible process,Therefore,the scientific research on the mechanism of disc degeneration has great significance at preventing cervical and lumbar spine disease.The research have been carried out from different aspects and is a concentrating subject of worldwide scholars in this area.Every new discovery of the mechanism of disc degeneration will be the premise and foundation to the development of the treatment.Animal model is a good way to study the occurrence of disc degeneration and test mechanism for the effectiveness of various treatment.The set up of a reliable lumbar disc degeneration animal model provides pre-conditions for the study of the pathogenesis of disc degeneration and also the test carrier for the study of new treatments.Chinese traditional medicine has embodied several advantages in the treatment of lumbar disc degeneration.Therefore,the possibility of medicine to the disc through the impact of cytokines to inhibit disc degeneration is a problem worthy of further investigation.The study of Chinese medicine to combat vertebral disease will deepen the research of traditional Chinese medicine.on the postponement of the degeneration of lumbar spine disease.This experiment is designed to study the function of Duhuo Jisheng Decoction of female Hartley guinea pigs to prevent the natural aging process of the role of disc degeneration and provide scientific theories and experiments basis for Chinese medicine in the prevention of disc degeneration from the perspective of modern medicine.Research purposesThrough this experiment,female Hartley guinea pigs natural aging lumbar intervertebral disc degeneration model,dynamic-month-old guinea pigs to observe the morphological changes of lumbar intervertebral disc,blood biochemical changes in the lumbar set up a reliable animal model of disc degeneration;observation Chinese medicine treatment of intervertebral disc degeneration in guinea pigs with blood cytokines and the effects of proteoglycan content changes Duhuo Jisheng preliminary study on guinea pig soup disc degeneration mechanism.Research MethodsSelect 104 female Hartley guinea pigs.Based on their ages,the pigs are randomly divided into 4 groups with 8 in every group,they are normal group, model group,treatment group(Duhuo Jisheng Decoction),positive control group(the Portuguese legislation).From lumbar intervertebral disc L4-5 for HE, Safranine O/fast green,Mallory staining to observe morphological changes of nucleus pulposus disc,testing of serum specimens in each group in the changes in proteoglycan content.Immunohistochemical detection of disc nucleus in IL-1β,TNF-α,MMP-1,MMP-3 content.Results1 Histomorphology:From HE,Safranine O/fast green,Mallory staining and were observed,with obvious morphological changes of old growth and the normal group,model group of the disc significantly reduced the number of notochord cells,myeloid concentric nuclear shrinkage,fibrosis,thinning of the cartilage end-plate defect even.Chinese medicine and western medicine group were observed in slices can also be found in a certain number of notochord cells,degeneration of nucleus pulposus than the model group. 2 Levels of blood biochemical:serum protein polysaccharides Elisa test results show that the model group,serum protein,polysaccharide content decreased with age,and differences between different-month-old group significantly.The treatment group and positive control group decreased the level of serum protein polysaccharides light than the model group,particularly in the treatment group the highest polysaccharide content of serum protein.3 The molecular level:The immunohistochemical staining method to observe IL-1β,TNF-α,MMP-1,MMP-3 in the degeneration of intervertebral disc tissue and content of change,growth and decline with age,different-month-old significant differences between groups,the model group of western medicine group medicine group express higher than normal group model has significant differences,and the model group,more evident on disc of traditional Chinese medicine on inflammatory factors in the degradation of the enzyme and there is a certain degree of inhibition increased role.Conclusion1 Through the model of Female Hartley guinea pigs'natural degeneration of aging,dynamic changes in the structure of intervertebral disc tissue can be observed,especially the nucleus pulposus morphology,cytokines and blood biochemical changes.It can better simulate the primary change in disc degeneration.2 Through the inhibition of inflammatory factor-degrading enzyme expression and increased proteoglycan content,Duhuo Jisheng Decoction of Chinese medicine can improve the biomechanical properties of intervertebral disc and adjust its microstructure.Finally achieve the objective of prevention and cure of disc degeneration.
