Objective:To find out haloperidol's toxicity to the rats'kidney,and investigate the protective effects of Huang Lian E Jiao He Ji(HLEJ) to the kidney and try to approach its mechanisms.Methods:Seventy-two SD rats were randomly divided into 6 groups:Bland Group, Toxic Group,Interfered Group,Low-dose Group,Middle-dose Group and High-dose Group.No intervention was given to the Bland Group.Rats in the Toxic Group were operated by HCRT.Enalapril and HCRT were given respectively to the Interfered Group.Beside HCRT,HLEJ was given respectively in low,middle and high dose to the Low-dose Group,Middle-dose Group,High-dose Group by gavage.After 30 days, urine of 24 hours were collected for the urinary routine test(URT),the al-microglobulin(a1MG),the micro albumin(MA),the kidney tissue were be taken for pathological test.Results:.Compared with the Blank Group,the increased level of the a1MG was observed(P<0.01).The largest alterative of the a1MG is the Toxic Group(67.5%).The next is the High-dose Group(47.5%),the third is the Interfered Group(35.0%).The Toxic Group compared with the Interfered Group and the High-dose Group,the increased level of the a1MG was observed(P<0.01).At the same time,the increased level of MA and a1MG in Bland Group were indistinct(P>0.05).Compared to the Blank Group a1MG in other groups all changed obviously(P<0.01).Compared with the Interfered Group and the High-dose Group,a1MG in the Toxic Group increased distinct(P<0.05).Although a1MG in the High-dose Group was higher than the Interfered Group,they have no statistical sense.a1MG in different dose group didn't have statistical sense.Kidney biopsy:The glomerulus didn't have evident lesions.But we can find some lesions in the renal tubulointerstitum,which was the most evident in the Toxic Group.Fibrosis was evident in the Toxic Group,and lighter in the High-does Group,but disappeard in the Interfered Group.Rats in the High-does Group have more serious inflammatory infiltration than the Interfered Group,but lighter tubular lesions.Conclusion:1.Large dosage haloperidol had the distinct renal toxicity,mainly displayed on the renal tubulointerstitum.2.HLEJ could decrease urine protein effectively,protect the renal function,and lessen the renal damage caused by the haloperidol.3.Patients who took the haloperidol for a long time could try to combine with HLEJ.Maybe It could protect the kidney function in early.
