The Relationship Between Inhibition AQP4 Expression And Brain Edema Of Cryptococcus Infection

Cryptococcus neoformans is one of the most important mandical fungus,which could infect both immunocompromised,and immumocompetent host.The incidence of cryptococcosis has increased significantly during the past decades.It may infect all the organs of the body with the increasing morbidity of immunosuppressive states such as acquired immunmdeficiency syndrome.The meningitis or meningoencephalitis caused by cryptoccus neoformans striking the Central Nervous System is a lifethreatening infection with huge mortality.Currently,to study on pathogenesis of brain edema of cryptococcosis is very important. Althougt the mechanism of the infection is rarely to known,some studies show that Cryptococcus neoformans may be inhaled as spore,crossing the air-blood barrier even the blood-brain barrier into the CNS.ObjectiveTo research the relationship between inhibition AQP4 expression and brain edema of cryptococcus infection,we established the cryptococcal meningitis model in Wistar rats. This work was to investigate the changes of brain edema of CNS cryptococcus infection after inhibition AQP4 expression,and to study the relationship between the brain edema and the changes of AQP4 expression.MethodsIn this research,we injected a short hairpin plasmid vector psilencer3.1 that can specifically suppress AQP4 expression.Wistar rats were divided into AQP4 RNA interference group and sham operation group,we injected recombinant plasmid into the right lateral cerebral ventricle with Lipofectamin 2000.After 7 days,we injected Cryptococcus via the right lateral cerebral ventricle.We used western blotting and dry-wet weight to observed the change of AQP4 and brain edema.ResulesAfter 7 days,we known AQP4 was reduced after AQP4 RNA interference.The peak time is from 7d to 14d.After injected cryptococcus,the brain water content was significantly higher in Cryptococcus infection group and AQP4 RNAi Cryptococcus infection group than that in sham operation group at 6h,12h,24h and48h,the peak time of it was at 12h, the AQP4 RNAi Cryptococcus infection group is higher than the test group.ConclusionFrom the resules of our research imply AQP4 is play an important role in the process of Cryptoccosis.To inhibition AQP4 expression can aggravated the brain edema of Cryptococcus infection.
