Objective:To investigate the effects of the folic acid on human cervical carcinoma cell line in vitro.Methods:â‘ MTT method used to detect the proliferation of Hela cells co-cultured with folic acid of different concentrations and time.â‘¡Detect the percentage of apoptosis by flow cytometry to 48h Hela cells cultured.â‘¢The mRNA expressions of C-myc and P16 and P53 genes were determined by reverse transcription polymerase chain reaction(RT-PCR).â‘£The protein levels of C-myc and P16 and P53 were assayed by Western blotting.Results:â‘ Folic acid at concentrations of 750 and 1875 nmol/L could inhibit the growth of 24h Hela cells cultured significantly(P<0.05);Folic acid at concentrations of 375,750 and 1875 nmol/L could inhibit the growth of 48,72h Hela cells cultured significantly(P<0.05);The inhibition rate was increased in the dose-effect and time-effect manners.â‘¡Cell apoptosis analysis showed that folic acid could make the Hela cell apoptosis rate increased significantly.â‘¢There was no significant different in the level of P16 mRNA expression between 18nmol/L folic aid groups and control groups,but the mRNA expressions of C-myc and P53 gene were significantly difference in all of folic acid groups than control group.â‘£There was significant different in the protein levels of C-myc and P16 and P53 among all of groups.Conclusion:â‘ Folic acid can inhibit the Hela cells growth in vitro.â‘¡Folic acid can induce Hela cells to apoptosis by its enhance C-myc or reduce P16 and P53 genes.
