Invasion and metastasis are the ma in causes responsible for failure of surgica loperation and death of patients in treating ma ligna nt tumors, relating to a seriesof very complicated interactions. Tumor cells detach from prima ry focuses firstlyand then adhere to Extracellular matrix (ECM). Degradation of Extracellularmatrix is a key step for metastasis. Furthermore, the loss of cell-cell adhesionpla ys an important role in the early stage of invasion and metastasis of tumorcells. In recent years,β-catenin (β-cat) has become a hotspot for study due to itsdouble role in the adhesion of cells and the signa l transaction in Wnt pathway.Meanwhile,ovaria n cancer is so apt to recurrence and metastasis that it is difficultto treat among gynecological tumors. To study the mecha nism of recurrence andmetastasis, and to improve the 5-year survival rate of patients,it is obliged to findout a new target in the treatment of ovaria n cancer.In this experiment, we observe the expression ofβ-cat in high and lowmetastatic huma n ovary serous cystadenoca rcinoma cells and investigate therelationship betweenβ-cat and the metastasis of ovaria n cancer. Besides, the characters of increased expression ofβ-cat in the treatment procedure usingEpirubicin were also studied so as to offer some experimental reference and gistfor clinica l applica tion.1. The expression ofβ-cat protein in ovaria n tumorsSpecimens come from obstetrics and gynaecology,pathologica l science of theFourth Medicine Military University . paraffin-embeddedtis- sues were tested byclinica l pathology and immunohistochemistry . One hundred and sixty-six caseswith complete clinica l and pathologica l data were analyzed,. The avera ge agesof the patients in these cases are 56.4 and before operation no any treatment wasused by them such as chemotherapy, radiotherapy and biologica l therapy .Tostudy the significa nce ofβ-cat expression in tumor transformation andprogression and its relationship with clinica l pathology characteristics. usingimmunohistochemistry SP staining method examine the expressions ofβ-cat in126 cases of ovaria n canc ers,20cases of borderline and ma ligna nt tumors,10cases of ovaria n benign tumors and 10 cases of norma l ovaries. They areanalyzed by Chi-square test of SPSS12.0.(1) Epithelia l cells of ovaria n tumours were strongly positive on membraneforβ-catenin with cytoplasmic positive expression ofβ-catenin .The positive rateofβ-cat expression in ovaria n cancers was significa ntly higher tha n that inborderline and ma ligna nt tumors ovaria n, benign tumors and norma l ovaries,(P〈0.01)(2) The expression ofβ-cat in ovaria n cancers was correla ted with histologicaltyping(P〈0.05). A positive correla tion existed between the levels ofβ-cat in thepatients afflict with ovaria n cancer and the stage..The expression ofβ-cat is87.5% in stageⅠcompare with 73.3% in stageⅣ.2. Study on relation betweenβ-cat and the metastasis of ovaria n cancer High and low metastatic huma n ovary serous cystadenoca rcinoma cell lineHO-8910PM and HO-8910 (reserved in laboratory of Fourth Military Medica lUniversity ) were chosen in the experiment, Western blotting was used todetected the protein levels ofβ-cat, The RNA levels ofβ-cat was detected by RTPCRmethod. The adhesion assay was used to detect the cancer cells adhesionabilities, the cell invasion and migration abilities were detected by Transwellchambers.Results: (1) Western blotting and RT-PCR detection showed the highexpression ofβ-cat HO-8910 but low expression in HO-8910PM. (2)β-catlocated in the cell membrane of HO-8910 with bright green fluorescence, butexpressed in the cytoplasm as well as the cell membrane of HO-8910PM withweak green fluorescence.(3) The invasive ability of HO-8910PM was greater thanthat of HO-8910, but their difference of migratory ability was not remarkable. (4)Cell adhesion assay. There were no remarkable difference of adhesion abilitybefore 20min and after 100min between the two cell lines, but assay showed thaton 20min and 40min the ability of HO-8910PM adhesion to extracellular matrixwas stronger than tha t of HO-8910.3. Characters of increased expression ofβ-cat in the treatment procedure usingEpirubicinEpirubicin is a kind of Anthracycline anticancer drugs. It can achieve tumorchemotherapy by inserting DNA base pairs to get degradation of DNA andinhibitory of DNA TopoⅡ. Proliferation inhibition and apoptosis induction ofEpirubicin were widely applied in treatment of ma ligna nt tumors. In this studyHO-8910 cell lines were divided into med icine control group (Epirubicin 5μmol/l)and norma l group, the expression ofβ-cat protein were tested by Western blotrespectively. Results: Expression ofβ-cat in experiment group were significa ntlyhigher than those in control group. With the increasing of Epirubicin, the expression ofβ-cat was enhanced. Conclusion:These results suggest that the highexpression ofβ-cat may have a close relation to the anti-tumor effects ofEpirubicin.
