Tryptanthrin(TRYP), chemical name is indole[2,1-b]quinazoline-6,12- dione , have a function of anti-inflammatory, anti-bacterial, anti-parasitic and anti-tumor activity of pharmacology. The investigate of TRPY in home is still under initial stage. There no relate reports about the effect of TYPR on the cardiovascular. In this study, using coronary artery ligation method cause the myocardial ischemia in rats , through investigate the cardiac function,cardiac pathology,myocardial mitochondrial ultrastructural from which effect of TRPY on the myocardial ischemia in rats . Using the method of hydroxyl ammonia,thiobarbituric acid and so on, to investigate the content of some enzymes, such as SOD,MDA and so on. Using immunohistochemistry investigate the protein content of myocardial tissue COX-2,identify the effect of TRYP on the cardiovascular system ,first discuss effect mechanism of TRYP on the myocardial ischemia。 1 Objective1.1 Observe the effect of TRYP on the cardiac function which from the coronary artery ligation in rats。1.2 Observe the effect of TRYP on the cardiac pathology,myocardial mitochondrial ultrastructural which from the coronary artery ligation in rats。1.3 Detect the SOD, MDA, LDH content of myocardial ischemia in rats .1.4 Observe the effect of TRYP on the COX-2 of ischemic myocardial tissue。1.5 Discuss the possible mechanism of the effect TRYP on the myocardial ischemia。2 Methods:2.1 Using the left anterior descending coronary artery ligation-induced myocardial ischemia model, then oral drug delivery . Using the overall experimental technique of rats physiology, observe the effect of TRYP on the cardiac function which from the coronary artery ligation in rats.2.2 Observe the effect of TRYP on the cardiac pathology,myocardial mitochondrial ultrastructural by light and electron microscopy .2.3 Using the method of hydroxyl ammonia,thiobarbituric acid,enzymatic,Nitrate reductase and so on, to investigate the content of SOD,MDA,LDH,NO,NOS,CK,TXB2,6-keto-PGF1α.2.4 Observe cardiac tissue expression of COX-2 on myocardial ischemia in rats by immunohistochemistry .3 Results3.1 Tryptanthrin of 0.10g/kg can significantly improved the heart function of big rats with myocardial ischemia, it can strengthen the myocardial contractility, improved myocardial compliance , shows the left ventricular systolic, the left ventricular pressure differential calculus±dp/dtmax have obviously increased, which has obvious difference with the model group(P<0.05).3.2 Light microscope and electron microscope shows that tryptanthrin of 0.10g/kg can significantly reduced the myocardial ischemia of big rat with myocardial degeneration, myocardial fibrosis and lymphocyte infiltration, also can reduce the extent of mitochondrial swelling.3.3 Compared the model group with the sham-operated group, MDA increased,SOD activity decreased, CK and LDH content increased, NO,NOS measured values lower; 6-Keto-PGF1αlevel increased, the ratio of TXB2/6-Keto-PGF1αincreased, compared with sham-operated group, significant difference shows in this group (P<0.05). TRYP0.10g/kg can decreased the content of MDA, CK, LDH, increased SOD activity, increased NO, NOS date, decreased TXB2,6-Keto-PGF1αin plasma and myocardial tissue, compared with model group, shows significant difference. (P﹤0.05).3.4 Tryptanthrin can significantly inhibit the expression of COX-2 in cardiac muscle, the inhibition gradually enhanced with the dose increasing.4 ConclusionsTryptanthrin of 0.10g/kg play a significant protection role on big rats with liagated coronary , which it lead myocardial ischemia.
