Partâ… Expression of VEGF and flk-1 receptor in vessel wall of Denervated skeletal muscle in ratsObjective To investigate the the pattern of expression of VEGF and fetal liver kinase-1(flk-1) receptor in the vessel walls of denervated rat skeletal muscles.Methods Twenty-seven male adult SD rats were randomly divided into normal control group ( n=3) and experimental group ( n = 24). Rats of the experimental group received neurotomy to bilateral sciatic nerve and the gastrocnemius muscles were harvested at 4 hours, 8 hours, 12 hours, 24 hours, 3 days, 5 days, 7 days, 14 days postoperatively. Immunohistochemical method and image analysis were used to study the expression of VEGF and flk-1 receptor in vessel walls of the muscles .Results In normal rats, VEGF was expressed in vessel walls of the gastrocnemius, while flk-1 was only expressed in endothelium of the vessel. After neurotomy, the expression of VEGF was down-regulated in the first 8 hours. It then reached a lower than normal level and persisted for 3 days. The expression of VEGF was up-regulated from the 3rd day to the 14th day after neurotomy while its expression level was still lower than that in the normal control group. In the first 12 hours after neurotomy, the expression of flk-1 showed a significant fluctuation of up-regulated and then down-regulated in endothelial cells. It then enhanced gradually from the 24th hour until the 14th day.Conclusions The results revealed the change of VEGF and flk-1 receptor expression in vessel walls of denervated skeletal muscles , which establishes the basis for further studies on the mechanism of its vascular degradation and treatment from microcirculation. Partâ…¡The protection effect of Egb on vascular of Denervated gastrocnemius muscle and its mechanismObjective To investigate the protection effect of Egb on vascular of denervated gastrocnemius muscle of rat and its mechanism.Methods Fifteen male adult SD rats were divided randomly into normal control group ( n=3),control group ( n=6) and experimental group ( n = 6). The rats of control and experimental group were received a neurotomy to bilateral sciatic nerve , then rats of these two groups were gavaged respectively with Egb (100 mg/(kg?d)) and isovolumic normal saline everyday. Gastrocnemius were harvested respectively at 1th and 3th week postoperatively. Immunohistochemical method and image analysis were used to detection the CFR of denervated gastrocnemius and the expression of VEGF,flk-1 receptor and HSP70 in vessel wall.Results In normal rats, VEGF,flk-1 and HSP70 were expressed in vessel wall of gastrocnemius, and flk-1 was expressed only in endothelial cell of vessel. 1 week and 3 weeks after neurotomy, CFR of experimental group was higher than that of control group; the expression of VEGF and flk-1 in vessel wall of control group and experimental group were lower than that of blank control group, and the expression of experimental group was lower than that of control group; the expression of HSP70 in vessel wall of control group and experimental group were higher than that of blank control group, and the expression of experimental group was higher than that of control group.Conclusion The results confirm the protection effect of Egb on vascular of denervated gastrocnemius muscle;and we deem that the mechanism of protection was not promoting the expression of VEGF and its receptor flk-1, but was correlated with the increasing of HSP70...
