Objective Esophageal carcinoma is one of the common malignant tumors in digestive system. Chinese national survey during 1990-1992 show that the death rate of esophageal carcinoma is 17.38/100, 00. South area of Hebei Province China is one of the highest incidence regions of esophageal cancer in the world, with a mortality rate of more than 100 per 100, 000 people per year. The treatment methods for esophageal cancer are esophagectomy, chemotheraphy and radiotheraphy. To investigate the anti-tumor effect of tanshinoneⅡA on the human esophageal cancer cell line Eca-109 and its mechanism, this study detected the effect of TanⅡA on proliferation of Eca-109 cells and expression of bcl-2 and bax.Methods1 Effects of TanⅡA on the proliferation of Eca-109 cells was measured by MTT colorimetric method.2 The expression of bcl-2, bax mRNA level in cultured Eca-109 was evaluated by RT-PCR on 72 hours.3 The bcl-2, bax protein level in cultured Eca-109 was evaluated by western-blot on 72 hours.4 Data were analyzed using SPSS13.0 statistical software. A level of P<0.05 was considered statistically significant.Results1 The results of MTT: MTT colorimetric method showed that the proliferation of Eca-109 cells can be inhibited by TanⅡA. The 0.5μg/ml TanⅡA treated group for 24 hours and 48 hours has no statistically significant difference compared with control group. After treated with TanⅡA, the OD values of the treated groups( 1~4μg/ml )decreased compared with control group (P <0.05), so the proliferation of Eca-109 cells can be inhibited by TanⅡA. Furthermore, with the increasing concentration of TanⅡA and prolonging of treating time, the OD values decreased gradually, which indicate that the inhabitation of the proliferation of Eca-109 cells by TanⅡA displays a dose-and-time-dependent manner .2 The results of PCR: Following the increasing of the concentration of TanⅡA ( 1~4μg/ml ) for 72 hours, expression of bcl-2 mRNA decreased gradually compared with control group, but bax mRNA was increased gradually, and there was statistically significant difference between control group and every treatment group (P <0.05). With the increasing concentration of TanⅡA, expression of bcl-2 mRNA were decreased gradually, but bax mRNA was increased gradually.3 The results of Western: Following the increasing of the concentration of TanⅡA ( 1~4μg/ml ) for 72 hours, expression of bcl-2 protein decreased gradually,but bax protein was increased gradually, and there was statistically significant difference between control group and every treatment group (P<0.05). By increasing of the concentration of TanⅡA, expression of bcl-2 protein were decreased gradually, but bax protein was increased gradually.Conclusion1 TanⅡA could inhibit the proliferation and induce the apoptosis of human esophageal carcinoma cell line Eca-109 in vitro with a dose-dependent and time-dependent pattern,which provide us a new way to cure the esophageal cancer.2 TanshinoneⅡA could inhibit the growth of human esophageal cancer cell line Eca-109 in a dose-and-time dependent manner. Its possible mechanism might relate to induce cell apoptosis, downregulate bcl-2 and upregulate bax gene expressions.3 TanshinoneⅡA could be a new prospective anti-esophageal cancer candidates. It is worthy of more in vestigation and exploiture.
