| This study took Camptotheca acuminata(Nyssaceae) as material,an indigenous species of China,also pilot study on distribution of Camptothecin and 10-hydroxycamptothecin in it. Based on the study mentioned above,extraction from Camptothecin(CPT)and 10-hydrxycamptothecin(HCPT) in seeds and leaves of Camptotheca acuminata were optimized by response surface methodology(RSM).Optimum technology parameters on seeds that 50%of ethanol volume fraction,3min of homogenate time and 7(mL:g) of liquor to material,while, optimum technology paramers on leaves that 70%of ethanol volume fraction,6min of homogenate time and 11(mL:g) of liquor to material.Exploring purification process of Camptothecin as organic solvent extraction,liquid-liquid extraction,recrystallization etc,and determined the optimal conditions of the experiments,finally in pilot magnification,the purity of CPT in extracum is 98%and the extraction yield of cpt is 0.0216%.Optimized separating method of HCPT from the remainder extracts for the production of CPT with macroporous resins adsorption-desorption.X-5 resin offers the best adsorption and desorption capacities for HCPT and the optimum conditions for adsorption were that pH was 5,40℃of temperature, 4BV/h of flow rate and 5BV of sample processing volume.The optimum conditions for desorption were that 10-90%of ethanol were used as solvents for gradient elution,30-60%of ethanol eluates were collected as final product which was concentrated.The flow rate was 4BV/h and the volume of elution was 2BV.The mid-test results show that the content of HCPT in the product increased to 7.19%,and the recovery yield was 82.1%.Optimization of recrystallization to purified HCPT,and its purity achieved 95%.This thesis is obtained for materials on seeds and leaves of Camptotheca acuminata,at the same time producing CPT and HCPT on mid-test.This technology is lower production costs,higher rates and highly pure,and effective utilization of CPT and HCPT in Camptotheca acuminata has guiding significance. |
