Objective: Nanobacteria is a novel apatite mineral-forming bacteria involved in the sclerosis. There were some research indicated that kidney stone has a intimate relationship with nanobacteria. Calculus dentalis is one of the symptoms of periodontitis, nanobacteria may be the etiological factor. The etiology of periodontal disease is still not very clear. There are lots of theories about pathogenesis, but none of them can elucidate the pathogenesis of periodontal disease alone and entirely. The theory of infection has been pay close attention to recently. The theory of inflammation has been supported by more and more research report. The purpose of this study is to discover the evidence of nanobacteria in the saliva of the patients of the periodontal disease and try to find out the relationship between nanobacteria and periodontal disease and to offer a new researchful field for the formation and pathogenesis of periodontal disease. Methods: 1. Separation and cultivation were performed in 30 saliva sample of patients of periodontal disease and 30 saliva sample without periodontal disease which is a control group. 2. The samples of the culture were identified by Dahl MeGec-Russel calcium-alizarin red staining. 3. The positive samples were checked by transmission electron microscopy.Results: 1. After about a four-week culture period, many samples were in clumps and started to attach to the bottom of the culture vessel, and white-colored precipitates could be seen in the cultures by the naked eye. 2.Dahl MeGec-Russel calcium-alizarin red staining and Gram staining were peformed in the cultural sample, we found that the nanobacteria predominantly appeared as clusters. 3. By the electron microscopy, we found that the nanobacteria revealed 100-380nm particles. They appeared bacilli or coccoid in shape, either as single particles or predominantly as clusters by attaching to each other with biofilm-like material. 4. The positive rate of nanobacteria was significantly higher in the the periodontal disease group than contrast group which is without periodontal disease(63.3% vs 10.0%). The difference between them is significant in statistics(P<0.05).Conclusion: 1.We found the nanobacteria in the saliva of the patients of periodontal disease. 2. Preliminary explored that nanobacteria may be involved in the the pathological mechanism of periodontal disease which should be further confirmed.
