Objective:To investigate the effect of smoking and smoking cessation on content of IKKβprotein in type 2 diabetes rats.Methods:84 six-weeks-old Wistar rats were randomly divided into 2 groups:normal(N,n=42) and diabetes(D,n=42).N and D groups were assigned randomly into control 8 weeks subgroup(NC8w,DC8w,n=7),control 12 weeks subgroup(NC12w, DC12w,n=7),smoking 8 weeks subgroup(NS8w,DS8w,n=7),smoking 12 weeks subgroup(NS12w,DS12w,n=7),smoking cessation 8 weeks subgroup (NSC8w,DSC8w,n=7) and smoking cessation 12 weeks subgroup(NSC12w, DSC12w,n=7).Smoking groups were given passive smoking twice daily for 8 or 12 weeks and smoking cessation groups giving up smoking for 4 weeks after 8 or 12 weeks smoking.Then used western blot method to observe the content of IKKβprotein,body weight,the feature of serum lipids and their correlated cytokine.Results:(1) 8 weeks,p-IKKβprotein content of each group had no significant difference(P>0.05).12 weeks,p-IKKβprotein content of diabetes smoking group was higher than the diabetes control group and smoking cessation group(P<0.01).p-IKKβprotein content of normal smoking group was higher than the normal control group and normal smoking cessation group(P<0.01).(2) Compared to the control group with the same age,diabetes smoking group showed a decline in body weight but had no statistical significance (P>0.05).There were no differences between diabetes smoking cessation eight weeks group and diabetes smoking twelve group in body weight(P>0.05).(3) compared with normal control group,FBG;FINS,IL-6,TNF-α,TG,TC,FFA of diabetic control group increased,HDL decreased significantly(P<0.05);12 weeks,FINS,IL-6,TNF-α,TG,TC,LDL,FFA of smoking group were higher (p<0.05),after quitting the above biochemical parameters such as blood lipids and inflammatory factors decline.There were no differences between Smoking cessation eight weeks group and the control group(P>0.05).Conclusions:IKKβprotein possibly participates the occurrence and development in diabetes.With the duration of IKKβprotein of smoking type 2 diabetes rats increased.Smoking may through inflammatory mechanisms lead to dyslipidemia and insulin resistance.IKKβprotein was reduced after smoking cessation,to improve dyslipidemia and insulin resistance.It is beneficial for improving dyslipidemia and insulin resistance.
