Objective:To determine the serum levels of interleukin-12(IL-12) and interferon-γ(IFN-γ) levels in childhood patients with different types of acute leukemia(AL). To analyze the relationship with their expression levels and the clinical prognosis index,the therapy effect,and to explore their clinical significance.Methods:The serum levels of IL-12 and IFN-γwere measured by sandwich enzyme-linked immunosorbent assay(ELISA) method in 38 cases of childhood AL,which included 30 cases of acute lymphoblastic leukemia(ALL) and 8 cases of acute non-lymphoblastic leukemia(ANLL),and 20 healthy children as control group.At the same time,the serum levels of IL-12 and IFN-γwere detected in 20 cases of childhood AL before and after induction treatment,as well as to analyzed the relationship with the levels to treatment effect and the clinical prognosis indexes was analyzed.Results:1.The serum levels of IL-12 and IFN-γwere 85.26±36.03pg/ml and 42.16±17.13pg/ml,respectively,in the new diagnoses AL group,which were significantly lower than those of control group(p<0.05).In the new diagnoses ALL and ANLL subgroups,the serum levels of IL-12 were 98.22±42.95pg/ml, 79.22±22.08pg/ml,and the serum levels of IFN-γwere 42.83±16.71pg/ml, 39.63±19.62pg/ml,respectively,which were also significantly lower than those of control group(p<0.05).But there were no significance between the new diagnoses ALL and ANLL subgroups(p>0.05).2.After induction treatment in 15 cases,the serum levels of IL-12(142.80±72.90pg/ml) and IFN-γ(107.80±42.49pg/ml) were increased in post-treatment complete remission(CR) group,which were significantly different compared to prior-treatment(80.00±18.83pg/ml,63.20±16.67pg/ml)(p<0.01);while the serum levels of IL-12 and IFN-γin post-treatment CR group were no significance compared to control group.The serum levels of IL-12(75.20±13.75pg/ml) and IFN-γ(38.60±6.43pg/ml) in 5 cases of post-treatment no remission(NR) group were lower than those of control group(p<0.05);while there were no difference compared to prior-treatment group(p>0.05).The serum levels of IL-12 and IFN-γin CR group were higher than those of NR group,and the differences were significant(p<0.05).3.According to the clinical sub-group with risk factor in the new diagnoses AL in childhood,the serum levels of IL-12 were 83.08±48.32pg/ml in High risk (HR) group,85.83±27.23pg/ml in middle risk(MR) group and 87.86±34.83pg/ml in low risk(LR) group,which were no significantly difference(p>0.05).There were significantly difference in the serum levels of IFN-γamong HR group (31.23±8.46pg/ml) and MR group(44.13±15.30pg/ml) and LR group (59.00±21.80pg/ml)(p<0.05).In the new diagnoses ALL,the serum levels of IL-12 in T-ALL group(110.71±45.90pg/ml) were no significantly difference compared to those in B-ALL group(81.92±30.20pg/ml)(p>0.05);and the serum levels of IFN-γin T-ALL group(40.00±9.27pg/ml) were no significantly difference compared to those in B-ALL group(45.17±20.25pg/ml),too(p>0.05).4.The serum levels of IL-12 and IFN-γin the new diagnoses AL were correlated with count of blast cells in bone marrow(r=-0.655,r=-0.878,p<0.01). The serum levels of IL-12 and IFN-γin the new diagnoses AL were irrelevant to sex,the count of white cells,red cells,platelets and the hemoglobin levels in peripheral blood(p>0.05).5.The serum levels of IL-12 in the new diagnoses AL were correlated with the serum levels of IFN-γ(r=0.735,p<0.01).Conclusion:1.The patients with childhood AL had the low level expression of IL-12 and IFN-γ,and there was increase of their expression in CR cases of post-treatment. It showed that IL-12 and IFN-7 might be relevant to the growth and multiplication of leukemia cell in childhood AL.2.The serum levels of IL-12 and IFN-γin the childhood AL were increased in post-treatment CR group.The serum levels of them in post-treatment NR group were lower than those of control group.It showed that the levels of IL-12 and IFN-γwere related to the development of the children AL.3.The serum levels of IL-12 and IFN-γin the childhood AL were related to the clinical treating effect and count of bone marrow blastic cells.So dynamically detecting the change of serum levels of IL-12 and IFN-γmight help to get further knowledge of the quantity of leukemia cells and the effect of treatment in childhood AL.4.The serum level of IFN-7 was significantly difference among HR group, MR group and LR group.
