Objective:The serum concentration of TPS,HER-2 protein,Anti-P53 and SF were detected by enzyme linked immunosorbent assay in the patients of breast cancer in Guangxi.To study the value of combined assay of the four tumour markers in diagnosis of breast cancer,investigate their relationships with clinical and pathological characteristics.The immunohistochemistry method was used to detect the expressions of TPS,HER-2 protein,Anti-P53 and SF in breast cancer,to investigate the relationship between serum concentration and tissues expression of the four maker.Methods:43 patients with breast cancer were studied,31 cases with benign breast diseases and 31 cases healthy people were randomly selected as control, The serum concentration of TPS,HER-2 protein,Anti-P53 and SF were detected by enzyme linked immunosorbent assay in these three group.Besides, 43 cases of breast cancer,31 cases of breast benign lesion tissues and 31 cases of breast normal tissues were selected in the same patients,the immunohistochemistry method was used to detect the expressions of TPS, HER-2 protein,Anti-P53 and SF in these tissues.Statistical analysis was performed by using SPSS 15.0 X~2 test,LSD-t test and Pearson correlations.Results:(1) The serum concentration of TPS in breast cancer group, benige lesion group and normal control are 25.51±10.36 ng/ml,16.78±15.09 ng/ml,16.42±11.83 ng/ml respectively;the serum concentration of HER-2 protein in three group are 11.24±5.68 ng/ml,6.85±2.31 ng/ml,6.58±2.35 ng/ml respectively;the serum concentration of Anti-P53 in three group are 9.22±5.37 U/L,8.55±5.71 U/L,7.59±3.66 U/L respectively;the serum concentration of SF in three group are 359.68±151.52 ng/ml,156.48±133.38 ng/ml,125.79±85.42 ng/ml respectively.The concentration of TPS,HER-2 protein,Anti-P53 and SF in breast cancer group were higher than those in non-cancer group,there were statistical significance in the concentration of TPS,HER-2 protein and SF(P<0.05).But there is no statistical significance between the results of benige and normal control.(2)The sensitivity rates of TPS,HER-2 protein and SF in breast cancer were 72.1%,76.7%and 90.7%respectively,the specific rates were 69.4%,71.0%and 80.6%respectively.In different combined assay,(HER-2 protein+SF) was the ideal type(the sensitivity rate was 97.7%,the specificity rate was 56.5%).(3)The concentration of four tumour markers in breast cancer group were positive correlation.(4) In breast cancer group,high serum TPS levels were significantly associated with younger,stageⅢ,Ⅳ,lymph node involvement,negativity of both estrogen and progesterone receptors;high serum HER-2 protein levels were significantly associated with stageⅢ,Ⅳ, lymph node involvement,negativity of both estrogen and progesterone receptors;high serum Anti-P53 levels were significantly associated with younger,stageⅢ,Ⅳ;serum SF levels were significantly associated with lymph node involvement,negativity of both estrogen and progesterone receptors,and there were statistical significance;(5) the expressions of TPS,HER-2 protein, P53 and SF in breast cancer tissues were higher than they in breast benign lesion tissues and breast normal tissues,the difference is statistical significance.There were positive correlation between the serum concentration and the expressions in breast cancer tissues of the four tumour markers.Conclusion:(1)The high serum expression of TPS,HER-2 protein, Anti-P53 and SF in breast cancer clue to they can be used as tumor marker for diagnosis of breast cancer.(2) The serum expression of TPS,HER-2 protein, and SF have value in diagnosis of breast cancer.In different combined assay of TPS,HER-2 protein and SF,(HER-2 protein+SF) was the ideal type,it shows good value in the diagnosis of breast cancer.(3)serum concentration of TPS, HER-2 protein,Anti-P53 and SF significantly correlates with age,size,clinic stage,lymph nodes metastasis,ER/PR,indicating the four markers are probably useful in evaluation and prognosis prediction of breast cancer.(4)There were positive correlation between the serum concentration and the expressions in breast cancer tissues of the four tumour markers,it shows monitor the serum concentration of the four marker is helpful to know their expressions in tissues.
