| Objective:To investigate the expressions of Nogo-A and its receptor NgR protein levels in the brain of experimental allergic/autoimmune encephalomyelitis(EAE) rats. The expressions of Nogo-A and receptor were observed after treated by methylprednisolone.Methods:The experimental EAE mould was established by subcutaneous administrating coarse myelin basic protein(MBP)and all the rats were divided into three groups as normal control group, complete Freund adjuvant (CFA) group and treatment group,respectively. The EAE rats were divided into control group and high dose(30mg/kg) as well as low dose(10mg/kg) mythylprednisolone(MP) treated group, respectively. 7days later, either high or low dose of was given via tail vein. Rats were sacrificed at two time points (6 and 18 hr) post vein injection. Optic chiasm with 2mm around tissue were harvested and process for further analysis. Morphology examination of rat brain cortex was done with H&E staining under light microscope. The expressions of Nogo-A and receptor (NgR protein) were detected by immunohistochemistry(IHC) staining on 4μm thickness of Formalin Fixed Paraffin-embedded(FFPE) and positive cells per each 0.36 cm2 area under 40X magnification were counted and compared between different groups.Results:1 Model establishment After be given MBP, 45% of Rat had been development EAE.The control group and CFA group are not invaded.2 H&E Under microscope with routine H&E staining,several features are showed,such as (1) inflammatory cell infiltration;(2)around the venula massive phlogocyte encirclement;(3)assumes the oversleeve type;(4)perivascular has escapes the myelinic membrane change, also the spongiocyte proliferation are formed as a result of the spongiocyte proliferation.3 Imunohistochemistry Nogo-A and receptor(NgR) were detected by IHC.Positive cells from EAE group are significant more than those of normal rats and CFA group(P<0.01).Also,a statistics significant difference exits between high dose and low dose of MP treatment group(P<0.05).Two-two comparing,besides EAE group and MP-L group(P>0.05),others all have statisticly significance(P<0.05)Conclusion:1 The EAE rat model could be successful1y set up with subcutaneous injection of coarse myelin basic protein;2 The expressions of Nogo-A and Ng Receptor from EAE group were increased;3 MP-H may depress the Nogo-A and NgR of the EAE rat brain tissue,and the mechanism of MP to MS may be related to this inhibitory effect. |
