Part I: Expression of NOGO-A and NG-R in AstrogliomaObjective To investigate the expression of Nogo-A and NG-R(Nogo-A Receptor)in normal human brain tissues and human astroglioma tissues,and explore the relationship between the malignant degree of astroglioma and Nogo-A and NG-R, and the correlation of them.Methods The expression of Nogo-A and NG-R in 8 cases normal human brain tissues and 37 cases astroglioma tissues were detected by immunohistochemical staining, semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blot method.Results Nogo-A and NG-R which was only little expressed in human normal tissues, significant presented in astroglioma cell membrane and cytoplasm and blood vessel endothelium. In human brain astroglioma, mean optical density value for Nogo-A and NG-R increased accordingly with the advancing pathological grades (P<0.05). There were a significant positive correlation between the expression of mRNA and protein of Nogo-A and NG-R(r=0.990,P<0.05;r=0.943,P<0.05).Conclusion The expression of Nogo-A and NG-R is closely related to the malignant degree of astroglioma, furthermore, there are inter-association between Nogo-A and NG-R expression. It indicated that Nogo-A and NG-R perhaps promote the formation and development of astroglioma together. Part II: Expression of NOGO-A and NG-R in OligodendrogliomaObjective To investigate the expression of Nogo-A and NG-R in normal human brain tissues and human oligodendroglioma tissues,and explore the role of Nogo-A and NG-R during the neoplasia of oligodendroglioma.Methods The expression of Nogo-A and NG-R in 8 cases human brain issues and 12 cases oligodendroglioma tissues were detected by immunohistochemical staining and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) .Results Nogo-A and NG-R which was little expressed in human normal brain issues, highly presented in oligodendroglioma cell.Conclusion The expression of Nogo-A and NG-R in oligodendroglioma cells significantly higher than in normal tissues, presumed that Nogo-A and NG-R perhaps promote the formation and development of oligodendroglioma.Part III: Expression of NOGO-A And NG-R inNeuroblastomaObjective To investigate the expression of Nogo-A and NG-R(Nogo-A Receptor)in normal human brain tissues and human neuroblastoma tissues,and discuss the relationship between the malignant degree of neuroblastoma and Nogo-A and NG-R.Methods The expression of Nogo-A and NG-R in 8 cases human brain issues and 35 cases human neuroblastoma tissues were detected by immunohistochemical staining.Results Nogo-A and NG-R which was little expressed in human normal brain issues, highly presented in neuroblastoma cell. In human neuroblastoma, mean optical density value for Nogo-A and NG-R increased accordingly with the advancing pathological grades.Conclusion The expression of Nogo-A and NG-R in oligodendroglioma cells significantly higher than in normal tissues, presumed that Nogo-A and NG-R perhaps promote the formation and development of oligodendroglioma.Part IV:A study of NOGO-A And NG-R expression in Primitive neuroectodermal tumorsObjective To investigate the expression of Nogo-A and NG-R in normal human brain tissues and human primitive neuroectodermal tumors tissues,and explore the role of Nogo-A and NG-R during the neoplasia of primitive neuroectodermal tumors.Methods The expression of Nogo-A and NG-R in 8 cases human brain issues and 15 cases primitive neuroectodermal tumors were detected by immunohistochemical staining.Results Nogo-A and NG-R was only little expressed in human normal brain issues. But in human primitive neuroectodermal tumors, mean optical density value for Nogo-A and NG-R was highly increased.Conclusion The expression of Nogo-A and NG-R in human primitive neuroectodermal tumors significantly higher than in normal tissues, presumed that Nogo-A and NG-R perhaps promote the formation and development of human primitive neuroectodermal tumors. Part I: Role of Nogo-A and NG-R in PC12 cells axon growthObjective To investigate the expression of NOGO-A and NG-R during the differentiation of archineuron cells, and explore the role of NOGO-A and NG-R during the growth of them.Methods PC12 cell, a rat's phaeochromocytoma cell line, was cultured and was induced by nerve growth factor(NGF). The expression of NOGO-A and NG-R in PC12 cell growth were detected by immunofluorescence staining, semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blot method. The proliferation and neurite extension of PC12 cells was observed by inverse microscopy.Results NOGO-A and NG-R was not expressed in undifferentiated PC12 cells. In PC 12 cells induced by NGF, with cell axon extention, the expression of NOGO-A and NG-R increased gradually(P<0.05).Conclusion During PC12 differentiate to sympathetic neuron, the expression of NOGO-A and NG-R increased gradually, presumed that NOGO-A and NG-R may enhance neurite growth during early development of neuron. Part II: The role of Nogo-A and NG-R in PC12 cells NeuroendocrineObjective To investigate the expression of NOGO-A and NG-R and dopamine secretion during the differentiation of archineuron cells, and explore the efect of NOGO-A and NG-R on neuroendocrine of nerve cells during the growth of them.Methods PC 12 cell was cultured and was induced by nerve growth factor (NGF). Dopamine (DA) level in the culture medium was detected on the 0, 1st, 3rd, 5th and 7th days by ELISA. The expression of NOGO-A and NG-R in PC12 cell growth were detected by immunofluorescence staining, semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Western blot method.Results There were little dopamine secretion in the culture medium of undifferentiation PC12 cells, but no NOGO-A and NG-R was detected in them. In the culture medium of differentiation PC12 cells the 1st, 3rd, 5th and 7th days, the density of dopamine secretion and the expression of NOGO-A and NG-R were all increasing gradually (P<0.05).Conclusion The expression of NOGO-A and NG-R increased during PC12 differentiate to sympathetic neuron as long as the dopamine (DA) level, indicated that NOGO-A and NG-R perhaps regulated neuroendocrine of nerve cells during the growth of them.
