Effects Of Inhaling Salmeterol-fluticasone On The Differentiation Of T-helper Lymphocytes Of Peripheral Blood And The Lung Function In Asthmatic Patients

Objective To investigate the effects of salmeterol-fluticasone on pulmonary function and the differentiation of T-helper lymphocytes in asthmatic patients .Methods 22 subjects with mild or moderate asthma were enrolled in the present study. All subjects inhaled salmeterol-fluticasone for one month based on the individual principle. The expression of IL-4, and IFN-γin serum in subjects were detected by ELISA method before and after treatment. Meanwhile the pulmonary function (FEV1%pred, FEV1/FVC, PEF%pred) were also measuredin all subjects.Results The expression of IL-4 in serum in subjects with asthma before treatment was significantly increased(144.460±30.158 vs 204.475±82.44 ,P<0.01), while the expression of IFN-γand the ratio between IFN-γand IL-4 were significantly decreased (29.750±8.206 vs 16.440±5.236 , P<0.01; 0.218±0.087 vs 0.099±0.058,P<0.01)when compared with that in control group.The expression of IL-4 in serum in asthma subjects after treatment was significantly decreased (204.475±82.44 vs 174.044±59.891,P<0.01), while the expression of IFN-γand the ratio between IFN-γand IL-4 were significantly increased (16.440±5.236 vs 18.973±5.642 , P<0.05; 0.099±0.058 vs 0.126±0.674,P<0.01) when compared with that in asthma subjects before treatment . But the expression of IL-4 in serum in asthmatic subjects after treatment was remarkably higher than healthy control group (144.460±30.158 vs 174.044±59.891,P<0.05) , also the expression of IFN-γand the ratio between IFN-γand IL-4 were lower than that in control group (29.750±8.206 vs 18.973±5.642 ,P<0.01; 0.218±0.087 vs 0.126±0.674,P<0.01).Correlation analysis showed that the expression of IFN-γnegatively correlated with the expression of IL-4 (r =-0.512, P<0.01).After the treatment, the levels of FEV1% pred,FEV1 /FVC and PEF%pred all significantly improved compared with before treatment (86.18±10.41 vs 69.30±15.50,76.93±7.45 vs 67.75±10.42,93.27±17.15 vs 67.42±19.57,P<0.01) .Conclusions The expression of IFN-γwas decreased and the expression of IL-4 was increased in asthmatic patients. It is obvious that the pulmonary function can be improved by inhaling salmeterol-fluticasone. Salmeterol-flutic- asone can promote the expression of IFN-γ, meanwhile can Inhibit the expression of IL-4 in asthmatic patients. The potential mechanism of inhaling salmeterol-fluticasone in asthmatic patients might be through correcting the disbalance of function existing in Th1/ Th2 .
