Fucoidans are sulfated polysaccharides located in the intercellular of brown algae and fucoidans from different species of algae have different chemical composition. Some are only simple sulfated polysaccharides composed of fucose, but also complex sulfated polysaccharides which contain several monosaccharides such as mannose, rhamnose, glucuronide, glucose, galactose, xylose, fucose. Previous study showed that FPS has good anticoagulant, antihyperlipidaemic, antitumor and anti-HIV activities, and has became the focus of natural drugs. The novel marine drug fucoidan which was studied by Institute of Oceanology, Chinese Academy of Sciences is used for the treatment of nephrotic syndrome and has a good effect on the early renal failure. The injection fucoidan in this paper is a low molecular weight FPS prepared with hydrogen peroxide-ascorbic acid (1:1) system degradation and agarose gel column classification, and has an unexpected effect on nephrosis treatment.FPS was degraded by hydrogen peroxide-ascorbic acid system and Fe2+-hydrogen peroxide-ascorbic acid system, imitated the oxidation-reduction reaction of polysaccharides in the process of plant growth and development. The reaction is mainly affected by the ratios and concentrations of the degradation reagents. The sulfate, uronic acid and fucose contents and infrared absorptions of the low molecular weight prepared with hydrogen peroxide-ascorbic acid and Fe2+-hydrogen peroxide-ascorbic acid degradation system have no significant difference with the raw materials which indicated that the chemical compositions and structural units of the polysaccharide were not modified in the degradation. The polydispersion index of the samples ranges from 1.49 to 1.77. These data demonstrated that methods applied in this experiment could result in intensively degraded products.For analysis of the monosaccharide composition in fucoidan, precolumn derivation HPLC method was developed in this paper. The fucoidan were hydrolyzed into monosaccharide with 2 mol·L-1 trifluoroacetic acid. With ribose as the internal standard, the monosaccharide derivatives obtained with 1-Phenyl-3-methyl-5 -pyrazolone(PMP) were separated by reverse-phase HPLC using a developed gradient elution process, and monitored by ultraviolet detector at 245nm. In the concentration range of 0.10 mmol·L-1~2.0 mmol·L-1, the peak area of every monosaccharide had a good linear relationship with its concentration(r2≥0.999); every monosaccharide derivatives had good reproducibility and stability except glucuronic acid; the average recoveries of mannose, rhamnose, glucuronic acid, glucose, galactose, xylose, fucose were respectively 86.2%,95.1%,62.5%,102.0%,94.8%,66.6%,105.1%. This method can be used for determination of certain monosaccharide contents in fucoidan such as fucose, galactose and glucose.Physical and chemical properties of the injection fucoidan including traits, identification, inspection and determination were studied in this paper. At the same time, forced degradation experiments were performed. The results of the experiments provide a basis for quality standard establishment and preparation research.
