Objectives: Ganoderma lucidum polysaccharides can enhance the body's specific and non-specific immune function,so it is considered to be a immunomodulator.It has a certain degree of anti-tumor effect.It has become important in health products raw materials. It was rarely reported that Ganoderma lucidum polysaccharides on the joint use of chemical drugs reduced tumor immune suppression. In this study,Ganoderma lucidum polysaccharides combined with interferon, as well as Ganoderma lucidum polysaccharides United DCN (decorin, DCN)are used in the S180 tumor-bearing mice. To investigate immunosuppressive factor secretion and regulation of immune function. To study Anti-tumor and immunomodulatory mechanism of Ganoderma lucidum polysaccharides combined with chemical drugs from molecular level,and provide theories for the development of anticancer drugs, define its anti-tumor mechanism and clinical treatment.Methods:1.we set up S180 entities tumor mice model. The mice were divided into four groups with random.. There were physiological saline(NS) group, GLP group,IFN-γ(IFN-γ)group,GLP combined with IFN-γ(GLP+ IFN-γ)group. They were treated by GLP and IFN-γ. The volume and weight of tumors were measured.The transformation induced by ConA of spleen lymphocytes and the level of VEGF in serum and the mRNA level of TGF-β1were detected and analysed.2. we set up S180 entities tumor mouse model. There were physiological saline ( NS ) group , GLP(GLP) group , pcDNA3.1(+)-DCN(DCN)group and pcDNA3.1(+)-DCN+GLP(DCN +GLP)group.They were treated by GLP and DCN. The volume of tumors were measured at the 5th day on alternate days. The transformation induced by ConA of spleen lymphocytes and the level of TGF-β1 and VEGF in serum were detected at the 14th day, meanwhile the percentage of CD4+CD25+ regulatory T cells in CD4+T cells was detected and analysed.Results: 1.Ganoderma lucidum polysaccharides United IFN-γThe cancers were inhibited in respective treatment group. The tumor inhibition rates of GLP, IFN, GLP combined with IFN-γgroup were 41.9%,43.6%,53.4%(P﹤0.05); The lymphocyte proliferation rates of different group were (53.87±4.76)% , (59.84±8.05)%, (67.92±7.45)% respectively(P﹤0.05); The quantity of VEGF in respective group was lower than in control group(793.25±64.23, 642.79±82.34, 546.67±76.43, 239.24±93.46)(P﹤0.05);meanwhile the mRNA level of TGF-β1 decreased markedly(1.15±0.22, 0.85±0.04, 0.87±0.03, 0.69±0.23)(P﹤0.05).2. Ganoderma lucidum polysaccharides united DCNTo compare with control group, tumor growth significantly slowed down (P﹤0.05). The lymphocyte proliferation rates of different group were(53.87±4.76)%,(59.84±8.05)%, (67.92±7.45)%;The quantity of TGF-β1 and VEGF in respective group was lower than in control group(2505.78±23.53, 2040.65±26.24, 1986.74±19.48,1594.24±29.61)P﹤0.05;(800.23±17.24, 639.83±9.52, 556.85±10.26, 334.37±17.15)P﹤0.05;Meanwhile the proportion of CD4+CD25+ regulatory T cells in CD4+Tcells was significantly lower than control group(22.17±3.07%, 17.46±2.14%, 4.07±3.37%,10.58±2.04%)(P<0.01).Conclusion: GLP,IFN-γand DCN gene could significantly inhibite the growth of tumor and enhance the immune system activity of mice bearing S180.They can depress the secretion of immunosuppressive molecules TGF-β1and VEGF from S180 tumor cells. GLP combined with IFN-γand GLP combined with DCN can promote it.
