A Study Of Genetic Polymorphisms Of STR Locus On The Y Chromosome DYS521 Among Shanxi Han Population

Objective To make more Y-STRs apply to forensic science and human genetics research, et al. We selected the candidate of Y-STR loci (DYS521) from human Y chromosome genomic DNA, and investigated the Y-STR allelic frequencies in Han ethnicity of Shanxi province in China. Discuss the possibility of applying it to forensic casework.Methods (1)EDTA-treated blood samples were collected from 120 healthy unrelated Han ethnicity of Shanxi province in China. The blood of 20 females were collected as negative control. Fifty father-son pairs, different male animals and different tissues of one corpse were analyzed. (2)The Genomic DNA was extracted via method of phenol-chloroform and PCR amplification.Analyze the PCR products by non-denaturing polyacrylamide gel electrophoresis followed by silver-staining.(3)Each allele of this Y-STR locus was sequenced and the allele ladders were constructed.Alleles of this Y-STR locus were nominated based on recommendations of the International Society of Forensic Genetics. (4)Collect and calculate the data.Result Our studies indicated that there were four alleles in DYS521 loci in Han ethnicity of Shanxi province named that DYS521*14,DYS521*15,DYS521*16,DYS521*17;In DYS521 locus,the allele frequencies ranged from 0.1250 to 0.5083,and the Gene Diversity vale is 0.6565,both discrimination power(DP) and probability of exclusion(PE) were 0.6565 in forensic medicine;No products were observed in 20 female blood,and no mutation was found in 50 father-son pairs.The typing of different tissues were identical.This loci were specific to the human species.Conclusion The DYS521 loci is highly genetic polymorphism and stability. The data obtained can be used in individual identification, paternity test and population genetics investigation for Han ethnic group in Shanxi.
