Objective To observe the effects of retinoic acid(RA) and fluvastatin(FLU) on radiation-induced lung injury.To evaluate the possibility of using RA together with FLU as a new therapy for pneumonia and pulmonary fibrosis induced by radiation.Methods 80 female SD rats were divided into 4 groups:Normal control(group C),radiated group(group R),treatment group(group T) and prevention group(group P). The latter 3 groups were radiated with linear accelerator at a dose of 15Gy after anesthesia. RA together with FLU was given to rats in treatment group(group T) at a dose of 10mg / (kg·d) by gastrogavaging from seven days before radiation until the end of the experiment. The day after the radiation,RA together with FLU was gastrogavaged at a dose of 10mg/ (kg·d) in group P.The rats in other groups were gastrogavaged with the same amount of saline.5 rats of each group were sacrificed respectively on day 5,15,30 and 60. Hematoxylin and eosin(H&E) stain and Van Gieson stain were performed on the lung tissue slices to observe changes in microstructure.Expression of TGF-β1 was measured by immunohistochemistry staining.The field of observation of immunohistochemistry statining was measured by a computerized image analysis system.The data were analyzed by univariate of general linear model test of SPSS13.0.Results TGF-β1 positive areas of lung tissue increased in Group R at day 5, 15,30and 60 compared with Group C(P<0.01).While decreased deeply in Group T and P (P<0.05),and decreased in Group P at day 15,30 compared with Group T(P<0.01).Conclusion 1,Rat thoracic irradiation 15Gy disposable shaping the success of the radiation-induced lung injury model.2,Radiation-induced pulmonary injury in rat lung tissue TGF-β1 were significantly increased,indicating that TGF-β1 involved in the occurrence of radiation-induced pulmonary injury.3,RA together with FLU may alleviate radiation induced pneumonia and inhibit the proliferation of collagenous fibers.Prevention group more effectively compared with treatment group.
