Objective:The animal experiments:To observe the effect of hTR ASODN on telomerase activity and apoptosis in mice with leukemia cells,To evaluta the feasibility of increasing the efficacy of radiation therapy.Clinical trials:To observe the effect of Shen qi Fu zheng parenteral solution(sqfzps)on activity of telomerase in patients with acute leukemia,in order to explore the mechanism for the treatment of acute leukemia.Methods:In the animal experiments to establish the mouse model of acute leukemia, mice with leukemia were randomly divided into four groups,A control group,B hTR ASODN,C radiation,D hTR ASODN combined with radiation;killed the mice after the end of experiment,activity of telomerase in mononuclear cell of bone in leukemia mice was detected by telomeric repeat amplification protoco(TRAP-PCR-ELISA),cell apoptosis rate was detected by AnnexinV-FITC and the use of PI double staining and flow cytometry before and after treatment;to observe the change of telomerase activity and cell apoptosis rate of each group.In the clinical trials thirty six patients with acute leukemia were randomly single-blind divided into two groups,patients in treatment group(n=18) were treated by combination of therapy of standard induced relieving and sqfzps,patients in control group(n=18) were only treated of therapy of standard induced relieving;telomerase activity and cell apoptosis rate in mononuclear cell of bone in patients was detected before and after the therapy,the detection method is the same as the animal experiments.Results:In the animal experiments the mouse modle of acute leukemia was established;successfully;we found that the telomerase activity of the mouse with acute leukemia was obviously increased than the normol mice;telomerase activity was suppressed significantly by hTR ASODN radiation,hTR ASODN combined with radiation;activity of telomerase and cell apoptosis rate in mononuclear cell of bone in leukemia mice were significantly changed in treatment group than that in control group(P<0.05),hTR ASODN combined with radiation have the best effect than other groups(P<0.05),hTR ASODN has the similar effects as radiation in the telomerase activity;radiation was beter than hTR ASODN in cell apoptisis rate;hTR ASODN combined with radiation can significantly Prolonged the acute leukemia mouse' survival time than other group(P<0.05);in the clinical trials there were twenty seven patients has the positive telomerase in thirty six;the positive rat was 75%,telomerase activity was 3.852±1.879;nineteen patients have the positive telomerase in ANLL,the positive rat was 73.1%,eight patients have the positive telomerase in ALL,the positive rat was 80.0%,the positive rat of telomerase was no significant difference in ANLL and ALL(P>0.05);the treatment group has fourteen patients,the positive rat was 77.8%,the telomerase activity was 3.88±1.979;the control group has thirteen patients,the positive rat was 72.2%, the telomerase activity was 3.823±2.009;the positive rat of telomerase was no significant difference in the two group;the telomerase activity was significantly decreased and the cell apoptisis rat was obviously increased in treatment group and control group after treatment(P<0.05);sqfzps combined with chemotherapy was better than chemotherapy(P<0.05);the complete remission rate and total effective rate were no significant difference in the two group (P>0.05 );the toxicity of chemotherapy drugs such as bone marrow suppression,nausea and vomiting were significantly reduced.Conclusion:The increased telomerase activity may be the one of the mechanism;both hTR ASODN and radiation are effective to treat mice with acute leukemia by decreasing activity of telomerase,hTR ASODN can increase the radiation sensitivity for acute leukemia.SQFZPS to treat the acute leukemia by reduce the telomerase activity in bone marrow cells from the patients with acute leukemia and promote the cell apoptisis,moreover,can improve the effect of chemotherapy and reduce adverse reactions of Chemotherapy drugs.
